摘要
为研究不同酵母菌株培养物对羔羊生长性能、免疫水平及抗氧化功能的影响,本实验将80只45日龄健康断奶羔羊随机分为4组,每组20只。对照组饲喂全混合日粮(TMR),实验组在饲喂TMR日粮基础上分别添加BC酵母菌株培养物(实验Ⅰ组),XR4酵母菌株培养物(实验Ⅱ组)及BC+XR4株复合酵母菌株培养物(实验Ⅲ组),添加量为30 g/d/只。实验期共90 d,在实验16 d注射A型口蹄疫病毒(FMDV)疫苗和羊痘疫苗。实验期间每天计算各组羊平均日采食量(ADFI),分别于实验期不同时间计算各组羊平均日增重(ADG)及料重比(F/G)。结果显示,实验61 d~90 d,3组实验组羔羊的ADFI均显著高于对照组(P<0.05),其中Ⅲ组ADFI最高。在0~90 d,各实验组的ADG均显著高于对照组(P<0.05),其中Ⅲ组的ADG最高,但与Ⅰ、Ⅱ组均无显著差异;实验Ⅰ、Ⅲ组的F/G显著低于对照组与实验Ⅱ组(P<0.05)。于实验期不同时间采血后采用间接ELISA测定各组羔羊血清中IL-1β、IL-4、IL-6、IL-10、转化生长因子-β1(TGF-β1)、IFN-γ、肿瘤坏死因子-α(TNF-α)的分泌水平,并采用生化试剂盒检测血清中溶菌酶(LZM)、酸性磷酸酶(ACP)、总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)、丙二醛(MDA)和过氧化氢酶(CAT)的含量。采用间接ELISA检测实验期不同时间各组羔羊血清中A型FMDV和羊痘疫苗抗体水平。结果显示:除FMDV和羊痘疫苗抗体水平在实验期30 d之后才与对照组差异显著外,其他各检测指标在60 d之后与对照组均差异显著。具体而言,与对照组相比,实验Ⅰ组羔羊血清中IL-1β、IL-4、TNF-α、CAT、GSH-Px、T-AOC及羊痘疫苗抗体水平均显著升高(P<0.05),实验Ⅱ组羔羊血清中IL-1β、IFN-γ、CAT、T-AOC及羊痘疫苗抗体水平均显著升高(P<0.05),实验Ⅲ组羔羊血清中IL-1β、IL-6、TNF-α、IFN-γ、LZM、CAT、GSH-Px、A型FMDV及羊痘疫苗抗体水平均显著升高(P<0.05)。其中实验Ⅲ组羔羊血清中A型FMDV抗体水平显著高于对照组和实验Ⅰ、Ⅱ组(P<0.05)。90 d时实验Ⅲ组羔羊血清中LZM的含量显著高于对照组,其余两组均与对照组无显著差异,MDA含量显著低于对照组(P<0.05)。整个实验过程中各组羔羊血清中IL-10、TGF-β1、CAP及T-SOD含量均无显著变化。上述结果表明,不同酵母菌株培养物均能显著提高羔羊的生长性能、免疫及抗氧化功能,其中BC酵母菌株在提高羔羊抗氧化功能方面优于XR4菌株与复合菌株培养物,而复合菌株培养物在提高羔羊A型FMDV和羊痘疫苗抗体效价方面优于单菌株培养物。本研究为不同酵母菌株培养物应用于肉羊生产实践中的研究提供参考依据。
To study the effects of different yeast strains culture on the growth performance,immune level and antioxidant function of lambs,8045-day-old healthy weaned lambs were randomly divided into 4 groups with 5 lambs in each group.The control group was fed with a total mixed ration(TMR),whereas the experimental groups were supplemented with BC yeast strain culture(experimental group I),XR4 yeast strain culture(experimental group II),and the compound yeast strain culture of BC+XR4 strains(experimental group III)on top of the TMR ration,respectively,at a dosage of 30g/d per lamb.The experimental lasted 90 days,and type A foot-and-mouth disease virus(FMDV)vaccine and sheep pox vaccine were inoculated on the 16th day of the experiment.The average daily feed intake(ADFI)of the lambs in each group was computed every day during the experiment.The lambs were weighed at different time points during the experimental period,and the average daily gain(ADG)and feed/gain ratio(F/G)of the lambs in each group were calculated.The results demonstrated that from day 61 to day 90 of the experiment,the ADFI of the lambs in the three experimental groups was significantly higher than that of the control group(P<0.05),and the ADFI of the group III was the highest from 0 to 90 days,the ADG of all experimental groups was markedly higher than that of the control group(P<0.05).Among them,group III had the highest ADG,but there was no significant difference compared to groups I and II.The F/G of experimental groups I and III was significantly lower than that of the control group and experimental group II(P<0.05).Blood samples were collected at various times during the experimental period,and the secretion levels of IL-1β,IL-4,IL-6,IL-10,transforming growth factor-β1(TGF-β1),IFN-γ,and tumor necrosis factor-α(TNF-α)in the serum of the lambs in each group were determined by indirect ELISA.The contents of lysozyme(LZM),acid phosphatase(ACP),total superoxide dismutase(T-SOD),glutathione peroxidase(GSH-Px),total antioxidant capacity(T-AOC),malondialdehyde(MDA)and catalase(CAT)in the serum were detected using biochemical kits.The antibody levels of type A FMDV and sheep pox vaccine in the serum of lambs in each group were measured by indirect ELISA at different times during the experimental period.The results indicated that,except for the antibody levels of FMDV and sheep pox vaccine,which were significantly different from the control group only after 30 days of the experimental period,the detection indexes were significantly different from the control after 60 days.Specifically,compared with the control group,the levels of IL-1β,IL-4,TNF-α,CAT,GSH-Px,T-AOC and sheep pox vaccine antibody in the serum of lambs in experimental group I were significantly elevated(P<0.05).The levels of IL-1β,IFN-γ,CAT,T-AOC and sheep pox vaccine antibody in the serum of lambs in experimental group II were significantly increased(P<0.05).The serum levels of IL-1β,IL-6,TNF-α,IFN-γ,LZM,CAT,GSH-Px,type A FMDV and sheep pox vaccine antibody in the serum of lambs in experimental group III were significantly increased(P<0.05).Among them,the antibody level of type A FMDV in the serum of lambs in experimental group III was significantly higher than that of the control group and experimental groups I and II(P<0.05).At 90 days,the content of LZM in the serum of lambs in experimental group III was significantly higher than that of the control group,while the other two groups had no significant differences from the control group,and the content of MDA was significantly lower than that of the control group(P<0.05).Throughout the entire experiment,there were no significant alterations in the contents of IL-10,TGF-β1,CAP and T-SOD in the serum of lambs in each group.The above results suggest that different yeast strain cultures can remarkably enhance the growth performance,immunity and antioxidant function of lambs.Among them,the BC yeast strain culture was more advantageous than the XR4 strain and the compound yeast strain culture in improving the antioxidant function of lambs,while the compound yeast strain culture was more superior to the single yeast strain culture in elevating the antibody titers of type A FMDV and sheep pox vaccine in lambs.This study provided a reference for the application of different yeast cultures in meat sheep production practice.
