摘要
目的探讨白藜芦醇(resveratrol,RSV)对缺氧/复氧(hypoxia/reoxygenation,H/R)诱导的H9c2心肌细胞凋亡的保护作用及机制。方法以H9c2心肌细胞为研究对象,建立H9c2心肌细胞H/R损伤模型,流式细胞术测定细胞凋亡;Western blot检测凋亡相关蛋白Bax、Bcl-2、cleaved caspase-3及JAK2/STAT3信号通路蛋白的表达水平;采用JAK2抑制剂AG490研究抑制JAK2/STAT3信号通路对RSV减轻H/R诱导的H9c2心肌细胞凋亡的影响。结果RSV可有效抑制H/R诱导的H9c2心肌细胞凋亡,并促进p-JAK2和p-STAT3蛋白表达,而AG490干预后明显拮抗RSV抑制H/R诱导的H9c2心肌细胞凋亡作用。结论RSV通过激活JAK2/STAT3信号通路,抑制H/R诱导的H9c2心肌细胞凋亡。
Aim To investigate the protective effects of resveratrol(RSV)on hypoxia/reoxygenation(H/R)-induced apoptosis in H9c2 cardiomyocytes and the underlying mechanisms.Methods A model of H/R was established in H9c2 cardiomyocytes.Cell apoptosis was determined by flow cytometry.The expressions of Bax,Bcl-2,cleaved caspase-3 and JAK2/STAT3 signaling pathway proteins were detected by Western blot.The inhibitor of JAK2,AG490,was used to assess the effect of RSV on H/R-induced apoptosis in H9c2 cardiomyocytes after inhibiting JAK2/STAT3 signaling pathway.Results RSV could effectively inhibit H/R-provoked cell apoptosis in H9c2 cardiomyocytes and increase the expression of p-JAK2 and p-STAT3.After AG490 intervention,the anti-apoptotic effect of RSV on H9c2 cardiomyocytes caused by H/R was significantly counteracted.Conclusions RSV can inhibit H/R-induced apoptosis in H9c2 cardiomyocytes through the activation of JAK2/STAT3 signaling pathway.
