不同来源鲜茧∕干茧生丝微量成分的组成特征

Constituent characteristics of trace components of fresh cured cocoon silk from different sources

摘要 利用高效液相色谱对不同来源的鲜茧/干茧生丝样品进行研究,分析了蚕种和产区等因素对鲜茧干茧生丝微量组分的影响。并以生丝微量组分的色谱峰面积作为变量因子,利用聚类分析方法研究了各因素对生丝微量组分含量的影响。结果表明:源于不同蚕种的鲜茧生丝的微量组分存在明显差异,而源于不同品种、产地和茧期蚕茧的干茧生丝微量组分的差异较小。相似度评价和系统聚类分析结果表明,蚕种对生丝的微量组分产生明显影响,而茧期和产地因素对生丝的微量组分影响较小。正交偏最小二乘判别分析结果表明,蚕种是对生丝微量组分影响最大的因素,鲜茧干茧生丝差异较大的组分主要为保留时间8.80~17.43 min的部分。该分析方法可用于易区分鲜茧生丝和干茧生丝,包括鲜茧生丝的产地、品种及茧期等身份信息,为鲜茧/干茧生丝的商业鉴别、身份溯源等提供借鉴与指导。 Compared with cured cocoon raw silk,raw silk prepared from fresh cocoons(referred to as fresh cocoon raw silk)has many problems in silk quality,such as higher sericin content and poor cohesion performance,which may be unfavorable to the quality of silk fabrics.In the past 10 years,there has been an urgent need of identification methods for identification of fresh cocoon raw silk and dried cocoon raw silk,but relevant detection technology is still lacking.There are only statistically significant differences in the structure and properties of fresh and dried cocoon raw silk,which makes it difficult to distinguish between the two raw silks by conventional test methods.The trace components in raw silk are closely related to factors such as the original production zone of cocoons,silkworm varieties,season of sericulture,and silk reeling process.Due to the difference in the processes of cocoon drying and cocoon cooking,there is theoretically a significant difference in the residual trace components in the raw silk spun from fresh cocoons and high-temperature cured cocoons.Analysis of the composition of trace components in the raw silks may be an effective method for distinguishing between the two types of raw silk.However,factors such as silkworm varieties,cocoon production areas,and season of sericulture may interfere with the differences in trace components between fresh cocoon raw silks and cured cocoon raw silks.Therefore,a large number of fresh cured cocoon raw silk samples from different sources were studied using high-performance liquid chromatography,and the effects of factors such as silkworm varieties and cocoon production areas on the trace components of fresh and cured cocoon raw silks were analyzed.And the peak area of liquid chromatography curves of the trace components in raw silk was used as a variable factor,the influence of various factors on the content of trace components in raw silk was studied using cluster analysis method.The results showed that there were significant differences in the trace components of fresh cocoon raw silk derived from different silkworm species,while the differences in trace components of cured cocoon raw silk derived from different varieties,original production zones,and sericulture seasons were relatively slight.The results of similarity evaluation and systematic clustering analysis indicate that silkworm varieties have a significant impact on the trace components of raw silk,while cocoon production seasons and production zone have a relatively slighter impact on the trace components of raw silk.The results of orthogonal partial least squares discriminant analysis indicate that silkworm eggs are the most influential factor on the trace components of raw silk,and the components with significant differences between fresh cocoon raw silks and cured cocoon raw silks are mainly those components with retention times ranging from 8.80 minutes to 17.43 minutes.This analysis method can be used to easily distinguish between fresh cocoon raw silk and cured cocoon raw silk,including identity information such as original production zones,silk varieties,and season of cocoon production of fresh cocoon raw silk.In this study,a method was established based on the analysis of the intrinsic difference components of fresh cocoon raw silk and dried cocoon raw silk,and on the basis of extensive sampling,liquid chromatography and statistical methods were used to classify and evaluate the similarity of samples,and the analysis results were accurate and reliable.This method could provide reference and guidance for commercial identification and identity tracing of fresh cocoon raw silks and cured cocoon raw silks.
