摘要
目的:基于生物信息学方法分析原发性高血压(EHT)的关键基因,预测潜在的治疗中药及靶向中药活性成分。方法:在GEO数据库选取GSE75940数据集,筛选EHT差异表达基因(DEGs)后,进行富集分析,构建蛋白互作网络,筛选关键基因。利用cBioPortal数据库对关键基因进行基因突变及生存分析,在Coremine Medical、CTD数据库中筛选潜在中药及有效活性成分。结果:筛选出1832个DEGs,富集结果主要与突触、神经肽、神经活性、G蛋白偶联受体、中性粒细胞迁移等相关。7个关键基因是表皮生长因子受体(EGFR)、趋化因子受体4(CXCR4)、白细胞介素17A(IL17A)、整合素αM(ITGAM)、细胞质蛋白-酪氨酸激酶(BTK)、阿片黑素促皮质激素原(POMC)、CD86。预测的潜在中药可分为清热解毒凉血、祛湿化痰、补肝益肾三大类,与EHT火、饮、虚的病机相符,中药有效成分包括白藜芦醇、木黄酮、槲皮素、姜黄素、辣椒素、鱼藤酮、紫杉醇等,与EHT发生发展均有一定联系。结论:本研究筛选出7个EHT关键基因,预测出其靶向中药及有效活性成分,可为后续研究提供新的生物标志物,促进中医药防治EHT。
Objective:To analyze the key genes of essential hypertension(EHT)based on bioinformatics and to predict the potential targeted Chinese medicines and traditional Chinese medicine(TCM)active ingredients.Methods:The GSE75940 dataset was selected from the GEO database,and after screening for EHT differentially expressed genes(DEGs),enrichment analyses were performed to construct protein-interaction networks and screen key genes.The cBioPortal database was utilized for mutation and survival analysis of key genes.Potential targeted Chinese medicines and TCM active ingredients were screened in Coremine Medical and CTD databases.Results:A total of 1832 DEGs were screened and the enrichment results were mainly related to synapses,neuropeptides,neuroactivity,G protein-coupled receptors,and neutrophil migration.The seven key genes were epidermal growth factor receptor(EGFR),CXC chemokine receptor 4(CXCR4),interleukin 17A(IL17A),integrin alpha M(ITGAM),Bruton tyrosine kinase(BTK),pro-opiomelanocortin(POMC),and CD86.The predicted potential Chinese medicines could be categorized into three main groups:clearing heat and detoxifying and cooling blood,dispelling dampness and phlegm,nourish the liver and kidneys,which was consistent with the pathogenesis of EHT fire,phlegm,and weakness.The active ingredients of TCM include resveratrol,genistein,quercetin,curcumin,capsaicin,rotenone,paclitaxel,etc.,which were related to the occurrence and development of EHT.Conclusion:In this study,7 key genes of EHT were screened and predicted to target TCM and effective active ingredients,which can provide new biomarkers for subsequent studies and promote the prevention and treatment of EHT in Chinese medicine.
