摘要
试验基于国家标准方法——高效液相色谱法(HPLC),优化流动相和洗脱条件,建立高效准确测定奶粉中L(+)-抗坏血酸含量的分析方法。试验以20 g/L偏磷酸溶液为提取剂,C_(18)柱进行分离,以0.1%草酸溶液为流动相,检测波长246 nm,流速1 mL/min,进样体积20μL,使用紫外可见双波长检测器进行测定。采用优化的测定方法得到,抗坏血酸在0~100.0μg/mL范围内线性良好(R^(2)=0.9999),空白加标回收率为99.12%~101.68%,精密度试验的相对标准偏差分别为1.18%,1.50%和1.18%,均符合相关标准要求。结果表明:该优化方法快速准确,灵敏度高,能够满足国家标准分析要求,适用于奶粉中抗坏血酸含量的测定。
This experiment is based on the national standard method——High-Performance Liquid Chromatography(HPLC),to optimize the mobile phase and elution conditions,establishing an accurate and efficient analytical method for determining the L(+)-ascorbic acid content in milk powder.A 20 g/L phosphoric acid solution was used as the extracting agent,with separation conducted using a C_(18)column.A 0.1%oxalic acid solution was employed as the mobile phase,with a detection wavelength set at 246 nm,a flow rate of 1 mL/min,and an injection volume of 20μL,utilizing a UV-visible dual-wavelength detector for measurement.The optimized method showed good linearity for ascorbic acid in the range of 0-100.0μg/mL(R^(2)=0.9999).The recovery rate from blank spiking ranged from 99.12%to 101.68%,and the relative standard deviations from precision tests were 1.18%,1.50%,and 1.18%,all complying with relevant standard requirements.The results indicate that this optimized method is rapid,accurate,and highly sensitive,capable of meeting national standard analysis requirements,and suitable for determining ascorbic acid content in milk powder.
