摘要
旨在考察如舒方提取物对女性经前综合征(PMS)的作用。将48只雌性SD大鼠随机分为模型组,阳性对照组(舒肝颗粒),如舒方提取物低、中、高剂量组,通过慢性束缚联合催产素诱导建立PMS模型,连续21 d灌胃给药。采用糖水偏好测试、ELISA、qRT-PCR、HE染色、扭体试验及Western blot检测大鼠行为及血清、组织中的生物标志物。结果显示:模型组大鼠表现出抑郁样行为,5-HT水平下降,NE、CORT水平升高,5-HTR2C mRNA表达降低,子宫组织出现病理性变化,PAG区GABA_(A)R亚单位蛋白表达增加。与模型组相比,如舒方各剂量组显著提高糖水偏好指数和血清5-HT水平,降低NE和CORT水平,改善子宫病理学表现及减少扭体次数,降低PAG区GABA_(A)Rα4、GABA_(A)Rβ1、GABA_(A)Rδ蛋白的表达,上调下丘脑5-HTR2C mRNA表达。如舒方通过调节GABA_(A)R和5-HTR2C的表达,改善PMS大鼠的抑郁行为,减轻经前腹痛。
This study aims to investigate the effects of Rushu formula extract on premenstrual syndrome(PMS).Forty-eight female SD rats were randomly divided into a model group,a positive control group(Shugan granules),and low,medium,and high-dose Rushu formula extract groups.A PMS model was induced by chronic restraint combined with oxytocin,and the rats were administered the treatments via gavage for 21 consecutive days.Sucrose preference tests,ELISA,qRT-PCR,HE staining,writhing tests,and Western blot analysis were used to evaluate behavioral,serum,and tissue biomarkers in the rats.The results showed the model group exhibited depressive-like behavior,decreased 5-HT levels,increased NE and CORT levels,downregulated 5-HTR2C mRNA expression,and pathological changes in uterine tissue,and increased expression of GABA_(A)R subunits in the PAG region.Compared with the model group,the Rushu formula groups significantly increased sucrose preference index and serum 5-HT levels,reduced NE and CORT levels,improved uterine pathology,reduced writhing,decreased GABA_(A)Rα4,GABA_(A)Rβ1,and GABA_(A)Rδprotein expression in the PAG region,and upregulated hypothalamic 5-HTR2C mRNA expression.Rushu formula alleviates depressive behavior and reduces premenstrual pain in PMS rats by regulating GABA_(A)R and 5-HTR2C expression.
