摘要
基于胶体金免疫层析法(colloidalgold immunochromatography assay,GICA),建立一种快速检测羊奶粉中啶虫脒残留的方法。通过优化标记抗体参数、羊抗鼠抗体稀释倍数、金标抗体用量等条件,制备胶体金免疫层析试纸条,并评价试纸条的检出限、灵敏度、假阳性率、假阴性率和交叉反应。结果表明,所制备的胶体金免疫层析试纸条可在15 min内检出羊奶中啶虫脒残留,检出限为0.8μg/L,折算成羊奶粉的检出限为7.2μg/kg;灵敏度≥98%、假阳性率≤2%、假阴性率≤2%;与吡虫啉的交叉反应率为2%,与噻虫胺、啶酰菌胺、过氧化氢、葡萄糖、尿素不发生交叉反应。该方法操作简单、检测速度快、成本低,适用于羊奶粉中啶虫脒残留的高通量现场快速检测。
Based on colloidal gold immunochromatography assay,a rapid method for the detection of acetamiprid residues in goat milk powder was established.The colloidal gold immunochromatographic test strips were prepared by optimizing the conditions,including parameters of antibody for labeling,dilution ratio of sheep anti-mouse anti-body and gold-labeled antibody doage.Under the optimal condition,the detection limit,sensitivity,false positive rate,false negative rate,as well as the cross-reactivity of the test strips were evaluated.The results showed that the prepared colloidal gold immunochromatographic test strips could detect acetamiprid in goat milk in 15 min,and the detection limit was 0.8μg/L,which translated into goat milk powder to 7.2μg/kg,with the sensitivity≥98%,false positive rate≤2%,false negative rate≤2%.The cross-reaction rate with imidacloprid was 2%,and there was no cross-reaction with clothianidin,boscalid,hydrogen peroxide,glucose and urea.The method is simple to operate,fast to detect and low in cost,which is suitable for the high-throughput on-site rapid detection of acetamiprid resi-dues in goat milk powder.
