硫酸铜对河川沙塘鳢急性毒性和相关酶活性的影响

Effect and Analysis of Copper Sulfate on Acute Toxicity and Metabolism-related Enzyme Activities of Odontobutis Potamophila

摘要 开展了硫酸铜(CuSO_(4))对河川沙塘鳢(Odontobutis potamophila)急性毒性和相关酶活性的影响试验。采取静水生物测试法,以CuSO_(4)最低致死浓度范围的最大值为起点,按倍比稀释法设定0.3、0.5、0.7、0.9 mg/L 4个浓度梯度,测定硫酸铜对河川沙塘鳢24 h、48 h、72 h、96 h的半致死浓度(LC50)和安全浓度(SC),以及鳃和肝组织中谷胱甘肽过氧化物酶(GSH-PX)、过氧化氢酶(CAT)、总超氧化物歧化酶(T-SOD)的活性。结果表明,24 h、48 h、72 h、96 h的LC_(50)分别为0.899、0.808、0.681、0.592 mg/L,96 h的SC为0.0592 mg/L。随着时间的增加和硫酸铜浓度的升高,鱼体内GSH-PX、CAT、SOD活性总体呈下降趋势。指出,河川沙塘鳢在不同浓度硫酸铜环境中的适应性和体内SOD、CAT、GSH-PX活性的变化,反映了河川沙塘鳢机体受到铜离子(Cu^(2+))的不同胁迫程度。 In order to investigate the acute toxic effect and influence of copper sulfate to Odontobutis potamophila,the whole process uses the static water bioassay method.According to the concentration and results set in the pre-experiment is,using the maximum of the minimum lethal concentration range as the starting point,four concentration gradients of 0.3,0.5,0.7 and 0.9 mg/L were set according to the dilution method.The activities of glutathione peroxidase(GSH-PX),catalase(CAT)and total superoxide dismutase(T-SOD)in gill and liver tissues were measured at 12 h,24 h,48 h,72 h and 96 h,respectively.The results showed that LC_(50) was 0.899,0.808,0.681,0.592 mg/L at 24 h,48 h,72 h and 96 h were respectively,and the safe concentration of CuSO_(4) was 0.0592 mg/L at 96 h.With the increase of time and CuSO_(4) concentration,the activities of GSH-PX,CAT and SOD in the fish generally showed a downward trend.The above results indicate the survival status and the changes of SOD,CAT and GSH-PX activities in the body of Odontobutis potamophila under different concentrations of CuSO_(4),which reflect the different degree of Cu^(2+)stress on the body of Odontobutis potamophila.
