摘要
为获得一种具有免疫功能的乳酸菌后生元,通过单因素和响应面试验优化乳酸菌后生元的制备工艺,通过斑马鱼试验验证该乳酸菌后生元对巨噬细胞吞噬功效的影响。结果表明,优化后制备乳酸菌后生元的最佳工艺条件为发酵温度33℃、发酵时间48 h、接种量5%,在此条件下胞外多糖含量为2303.18μg/mL。斑马鱼试验结果发现,乳酸菌后生元的最大检测浓度(MTC)为500μg/mL。不同浓度的乳酸菌后生元均能促进巨噬细胞吞噬功效,当乳酸菌后生元浓度达到250μg/mL时与模型对照组相比差异显著(P<0.01),浓度达到500μg/mL时效果与15μg/mL百令胶囊相当。研究结果表明乳酸菌后生元具有较强的提高免疫力功效。
In order to obtain postbiotics of lactic acid bacteria with immunity function,the production technology of postbiotics of lactic acid bacteria was optimized by single factor and response surface tests,and then the effect of lactic acid bacteria postbiotics on phagocytosis efficacy of macrophages was verified by zebrafish test.The results showed that the optimized best conditions for producing postbiotics of lactic acid bacteria were as follows:fermentation temperature 33℃,fermentation time 48 h,inoculum 5%,under these conditions,the content of extracellular polysaccharide was 2303.18μg/mL.The results of zebrafish test showed that the maximum test concentration(MTC)of lactic acid bacteria postbiotics was 500μg/mL.All different concentrations of lactic acid bacteria postbiotics could promote the phagocytosis efficacy of macrophages,and the difference was significant compared to the model control group when the concentration reached 250μg/mL(P<0.01),and the effect was similar to that of 15μg/mL bailing capsule when the concentration reached 500μg/mL.The results showed that the postbiotics of lactic acid bacteria had a strong effect on improving immunity.
