脂联素基因启动子区SNPs rs266729和rs17300539与TyG指数及冠心病的相关性研究

Study on correlation between SNPs rs266729 and rs17300539 in promoter region of adiponectin gene and index and coronary heart disease

摘要 目的 探讨脂联素基因启动子区单核苷酸多态性(SNPs)rs266729和rs17300539与TyG指数及冠心病(CHD)的相关性。方法 选择2018年1月—2021年6月本院心内科收治的238例患者作为研究对象,其中冠心病组139例,对照组99例。常规完善相关生化指标,同时采集两组患者外周血样本提取基因组DNA,采用聚合酶链反应(PCR)检测脂联素基因(ADIPOQ)的SNPs位点多态性。结果 冠心病组ADIPOQ rs266729和rs17300539与TyG指数无关,差异无统计学意义(P>0.05);两组间比较,ADIPOQ rs266729基因型频率和等位基因频率无统计学意义(P>0.05),ADIPOQ rs17300539基因型频率和等位基因频率差异有统计学意义(P<0.05);按照性别分层后,男性人群中rs17300539基因型分布频率在两组间有明显差异(P<0.05),多元回归分析表明rs17300539位点上的G等位基因是男性冠心病的独立危险因素之一。结论 ADIPOQ rs17300539位点上的G等位基因对于冠心病的发生具有性别特异性,是男性冠心病的独立危险因素之一,而对女性冠心病的发生未见影响。 Objective To investigate the correlation between single nucleotide polymorphisms(SNPs)rs266729 and rs17300539 in the promoter region of adiponectin gene and TyG index and coronary heart disease(CHD).Methods A total of 238 patients were selected from the Department of Cardiology,Jinchang City People's Hospital from January 2018 to June 2021,including 139 patients in the coronary heart disease group and 99 patients in the control group.Routine improvement of relevant biochemical indicators was conducted,and genomic DNA was extracted from peripheral blood samples of patients in both groups.Polymerase chain reaction(PCR)was used to detect SNPs polymorphism of adiponectin gene(ADIPOQ).Results There was no significant difference between ADIPOQ rs266729 and rs17300539 and TyG index in the CHD group(P>0.05).There were no significant differences in ADIPOQ rs266729 genotype frequency and allele frequency between the two groups(P>0.05).ADIPOQ rs17300539 genotype frequency and allele frequency were significantly different(P<0.05).The frequency of rs17300539 genotype distribution in the male population was significantly different between the two groups(P<0.05)after stratification by gender.Multiple regression analysis indicated that the G allele at the rs17300539 locus was an independent risk factor for coronary heart disease in men.Conclusions The G allele of ADIPOQ rs17300539 is sex-specific for the occurrence of coronary heart disease;it is one of the independent risk factors for CHD in men but has no effect on the occurrence of CHD in women.
