摘要
目的:探讨胃癌患者组织MutS同源物6(MSH6)、减数分裂后分离增强基因2(PMS2)表达水平与幽门螺杆菌(HP)感染及患者预后的相关性。方法:选取胃癌患者52例,收集术中切除的胃癌组织及癌旁组织(距肿瘤边缘<2 cm)。采用13 C和14 C呼气试验检测HP感染情况,记录HP阳性参考值(DOB),根据检测结果将患者分为HP感染组(29例)与HP未感染组(23例)。采用实时荧光定量PCR(RT-qPCR)检测胃癌组织MSH6、PMS2 mRNA表达,分析两者与临床病理特征的关系。采用Logistic回归分析患者预后的影响因素。采用Kaplan-Meier曲线分析MSH6、PMS2 mRNA表达水平与胃癌患者预后的关系。结果:胃癌组织MSH6、PMS2 mRNA表达水平明显低于癌旁组织,且两者表达水平与血管侵犯、TNM分期有关(均P<0.05)。HP感染组胃癌组织MSH6、PMS2 mRNA表达水平明显低于HP未感染组(均P<0.05)。血管侵犯、TNM分期Ⅲ-Ⅳ期及MSH6、PMS2 mRNA表达水平降低为胃癌患者预后的危险因素(均P<0.05)。MSH6、PMS2高表达患者3年生存率高于低表达患者(均P<0.05)。结论:胃癌组织MSH6、PMS2 mRNA表达降低,与患者HP感染密切相关。MSH6、PMS2高表达患者3年生存率较高。
Objective:To investigate the correlation of MutS homologue 6(MSH6)and postmeiotic segregation increased 2(PMS2)expression with Helicobacter pylori(HP)infection and prognosis of gastric cancer.Methods:52 patients with gastric cancer were selected as the research objects.Gastric cancer tissues and adjacent tissues(less than 2 cm from the edge of the tumor)resected during operation were collected.The 13 C and 14 C breath tests were used to detect HP infection,and the HP positive reference value(DOB)was recorded.According to the test results,the patients were divided into HP infection group(29 cases)and HP non-infection group(23 cases).The expressions of MSH6,PMS2 mRNA in gastric cancer tissues were detected by RT-qPCR,and the relationship between them and clinicopathological characteristics was analyzed.Logistic regression was used to analyze the prognostic factors of gastric cancer patients.The relationship between the expression levels of MSH6,PMS2 mRNA and the prognosis of patients was analyzed.Results:Compared with adjacent tissues,the expressions of MSH6,PMS2 mRNA in gastric cancer tissues were lower,and they were related to vascular invasion and TNM stage(both P<0.05).The expressions of MSH6,PMS2 mRNA in gastric cancer tissues in HP infection group were significantly lower than those in HP non-infection group(both P<0.05).Vascular invasion,TNM stageⅢ-Ⅳand the decreased expression of MSH6 and PMS2 mRNA were risk factors for the prognosis of gastric cancer patients(all P<0.05).Compared with low expression of MSH6 and PMS2,the 3-year survival rate of patients with high expression were higher(both P<0.05).Conclusion:The expressions of MSH6,PMS2 mRNA in gastric cancer tissues are decreased,which are closely related to HP infection in gastric cancer patients.The 3-year survival rate of patients with high expression of MSH6 and PMS2 was higher.
