饲料原料中沙门氏菌LAMP快速检测方法的建立

Establishment of rapid detection method for Salmonella based on LAMP in feed raw materials

摘要 我国现行标准GB13018—2017《饲料卫生标准》规定饲料产品中不得检出沙门氏菌。常规检验沙门氏菌的方法耗时长,对操作人员专业性要求高,导致企业生产成本增加,因此有必要开发一种具有独立知识产权的快速检测沙门氏菌的方法,用于产品内部质量控制。本研究建立一种快速检测饲料沙门氏菌的环介导等温扩增方法(LAMP),经条件优化后,建立的方法能够对沙门氏菌进行特异性扩增,灵敏度为1×10^(2) CFU,检测全程仅需约8 h,比常规检测方法快2~3 d,双盲试验结果表明,建立的方法与国家标准检测沙门氏菌的方法的符合率为100%,能够满足饲料厂家产品内部质控的需求。本研究能够为工业生产中沙门氏菌的快速检测提供技术支持。 Salmonella should not be detected in feed products in current standard(GB13018-2017)in China.The method of routine testing for Salmonella is time-consuming and requires high professionalism from operators,resulting in increased pro-duction costs for enterprises,therefore,it is necessary to develop a rapid method for detecting Salmonella with independent intellectual property rights for internal quality control of products.This study established a rapid detection method for Salmonella in feed using loop mediated isothermal amplification(LAMP).After optimizing the conditions,the established method can specifically amplify Salmonella with a sensitivity of 1×10^(2) CFU.The detection process only took about 8 hours,which was 2-3 days faster than conventional detection methods.The results of the double-blind experiment showed that the established method had a 100%compliance rate with the national standard for detecting Salmonella,and could be used for the internal quality control needs of feed manufacturers products.This study can provide technical support for the rapid de-tection of Salmonella in industrial production.
