摘要
目的研究姜黄素对成骨分化过程的潜在调控机制。方法通过网络药理学方法获取姜黄素调控成骨分化的靶点基因,构建蛋白质相互作用(PPI)网络,并进行GO、KEGG富集分析及分子对接处理。结果筛选出姜黄素调控成骨分化的靶点基因92个,主要涉及细胞凋亡,细胞凋亡负向调控及肌细胞增殖等生物过程;突触、膜筏及细胞连接等细胞成分;磷酸酶结合,蛋白激酶结合及酶结合等分子功能。靶点基因PRKACA、GSK3β、EP300在Wnt/β-catenin信号通路中处于重要位置,且均可以与姜黄素良好对接。结论姜黄素可以通过PRKACA、GSK3β、EP300调控Wnt/β-catenin信号通路来调控成骨分化过程。
Objective To investigate the potential regulatory mechanism of curcumin on the process of osteogenic differentiation.Methods The target genes regulated by curcumin on osteogenic dfferentiation were obtained through network pharmacology methods,and a protein protein interaction network(PPI)was constructed.Gene Ontology(CO),Kyoto Encyclopedia of Genes and Genomes(KECG)enrichment analysis,and molecular docking treatment were performed.Results A total of 92 target genes were screened for curcumin regulation of osteogenic differentiation,mainly involving biological processes such as cell apoptosis,negative regulation of cell apoptosis,and muscle cell proliferation,cellular components such as synapses,membrane rafts,and cell junctions.Phosphatase binding,molecular functions such as protein kinase binding and enzyme binding.Target genes PRKACA,CSK3βand EP300 in Wnt/βcatenin signaling pathway plays an important role and can be well docked with curcumin.ConclusionnCurcumin canpass throughPRKACA,GSK3βand EP300 regulates Wnt/β-catenin signaling pathway regulates the process of osteogenic differentiation.
