摘要
目的基于血清代谢组学探讨桂仙合剂治疗绝经综合征潮热大鼠的机制。方法将32只雌性SD大鼠随机分为对照组、模型组、克龄蒙组和桂仙合剂组,每组8只。除对照组外,其余组大鼠采用去卵巢手术建立绝经综合征潮热模型。证实造模成功后,桂仙合剂组给予33.3 g/kg桂仙合剂药液灌胃,连续30 d;克龄蒙组前3 d给予0.4 mg/kg戊酸雌二醇溶液灌胃,后2 d给予0.6 mg/kg戊酸雌二醇合醋酸环丙孕酮溶液灌胃,如此连续重复6次;对照组和模型组给予生理盐水灌胃,连续30 d。使用超高效液相色谱-四极杆/静电场轨道阱高分辨质谱(UPLC-Q-Orbitrap HRMS)技术对大鼠血清代谢物进行分析,并筛选潜在生物标志物及相关代谢通路。结果根据血清代谢组学结果,将模型组分别与对照组、桂仙合剂组比较,筛选出3种共同的差异生物标志物,分别为4β-甲基酶甾醇-4-甲醛、β-咔啉-3-羧酸、3-羟基-L-鸟氨酸;涉及5条共同的代谢通路,分别为精氨酸生物合成、组氨酸代谢、嘌呤代谢、类固醇激素生物合成、甘油磷脂代谢,其中甘油磷脂代谢是共同的差异代谢通路(P<0.05),而精氨酸生物合成是影响值最高的代谢通路(Impact=0.228)。结论桂仙合剂可能通过调节氨基酸和脂质代谢保护线粒体与血管内皮功能,从而改善绝经综合征潮热。
Objective It is to explore the mechanism of Gui Xian Decoction(GXD)in the treatment of menopausal syndrome with vasomotor symptoms(MPS-VMS)in ovariectomized rats based on serum metabolomics.Methods Thirty-two female SD rats were randomly divided into control group,model group,Climen group and GXD group,with 8 rats in each group.The rats of all groups except for the control group were subjected to ovariectomy to establish the models of MPS-VMS.After successful modeling,the GXD group was continuously treated with GXD 33.3 g/kg by gavage for 30 days;the Climen group was treated with estradiol valerate solution 0.4 mg/kg by gavage for the first 3 days and with estradiol valerate+cyproterone acetate solution 0.6 mg/kg by gavage for the next 2 days,and this therapy was repeated for 6 times;the control group and model group were treated with normal saline by gavage for 30 days.The serum metabolites in the rats were analyzed ultra high performance liquid chromatography quadrupole mass spectrometry(UPLC-Q-Orbitrap HRMS)technology and the potential biomarkers and related metabolic pathways were screened out.Results According to the results of serum metabolomics,the model group was compared with the control group and GXD group respectively,3 common differential biomarkers were screened out,named:4beta methylmasterol-4-carbaldehide,β-Carboline-3 carboxylic acid,3-hydroxy-L ornithine;it involved 5 common metabolic pathways:arginine biosynthesis,histidine metabolism,purine metabolism,steroid hormone biosynthesis and glycerophospholipid metabolism,in which glycerophospholipid metabolism was a common differential metabolic pathway(P<0.05),while arginine synthesis was the metabolic pathway with the highest impact value(Impact=0.228).Conclusion Gui Xian Decoction can improve menopausal syndrome with vasomotor symptoms via regulating amino acid and lipid metabolism to protect mitochondrial and vascular endothelial function.
