山楂饮片HPLC特征图谱及其基原区分研究 被引量:1

摘要 目的建立山楂饮片的高效液相(HPLC)特征图谱,并对不同基原的山楂饮片进行区分。方法以Agilent Eclipse Plus Rapid Resolution HD C_(18)(2.1 mm×100 mm,1.8μm)为色谱柱,以乙腈-0.1%磷酸溶液进行梯度洗脱,柱温为30℃,流速为0.3 mL/min,检测波长为320 nm。建立了山楂饮片特征图谱测定方法,利用聚类分析、偏最小二乘判别分析对不同基原山楂进行化学模式识别。结果山楂特征图谱共有17个共有峰,利用对照品指认出峰2为5-羟甲基糠醛、峰3为新绿原酸、峰4为绿原酸、峰6为隐绿原酸、峰13为金丝桃苷和峰14为异槲皮苷。聚类分析结果显示不同基原山楂分别聚集,PLS-DA显示色谱峰17、3(新绿原酸)、4(绿原酸)、9、10、15、16为不同基原山楂差异性成分,相对峰面积可以区分山楂、山里红、南山楂基原饮片。结论建立的特征图谱具有良好的重现性,化学模式识别和相对峰面积可用于区分不同基原的山楂。 Objective To distinguish hawthorn with different origins,and establish the HPLC characteristic fingerprint for hawthorn.Methods An Agilent Eclipse Plus Rapid Resolution HD C_(18)(2.1 mm×100 mm,1.8μm)column was used with a mobile phase of 0.1%phosphoric acid aqueous solution/acetonitrile(V/V)which used gradient elution at a flow of 0.3 mL/min.The detection wavelength was 320 nm and column temperature was set at 30℃.Different origins of hawthorn was identified in the fingerprint method by Hierarchical Clustering analysis and PLS-DA.Results The fingerprint of hawthorn had 17 common peaks.The peak 2 was 5-hydroxymethyl-2-furaldehyde,peak 3 was neochlorogenic acid,peak 4 was chlorogenic acid,peak 6 was cryptochlorogenic acid,peak 13 was hyperoside and peak 14 was isoquercitrin.Hierarchical Clustering analysis showed that different original hawthorns were clustered separately,and PLS-DA showed that peak 17,3(Neochlorogenic acid),4(Chlorogenic acid),9,10,15 and 16 were differential components of different original hawthorn,and the relative peak area could distinguish different hawthorn between Crataegus pinnatifida Bge.,Crataegus pinnatifida Bge.var.major N.E.Br.,and Crataegus cuneata Sieb.et Zucc.Conclusion The fingerprint is good reproducibility,which can be used to distinguish hawthorn from different origins by chemical pattern recognition and the relative peak area.
