摘要
细胞分裂素氧化脱氢酶(cytokinin oxidase/dehydrogenase,CKX)作为植物体内唯一能够通过催化细胞分裂素不饱和侧链裂解从而不可逆地降解内源性细胞分裂素的关键酶,在植物参与逆境胁迫过程中扮演着至关重要的角色。为研究其在荷花(Nelumbo nucifera)中的潜在功能,采用生物信息学方法鉴定了荷花CKX基因家族成员,使用q RT-PCR技术探讨了Nn CKXs在荷花不同组织和非生物胁迫条件下的表达模式。结果显示,9个NnCKXs分为4个亚组,其启动子区有大量响应非生物逆境胁迫和植物激素的顺式作用元件,且均有与异戊烯基腺嘌呤(isopentenyl adenine,iP)结合的活性作用位点。NnCKXs在不同组织中均有表达,表达量差异较大,在根和叶中表达量较高,且呈现对低温、盐害、厌氧胁迫的不同程度响应,并受逆境相关植物生长调节剂(脱落酸和茉莉酸甲酯)和生长类植物生长调节剂(生长素、赤霉素和细胞分裂素)的诱导表达。具体表现为NnCKX2、3、5、6和9表达量至少在两种非生物胁迫中均显著上调,而NnCKX1、4、7和8表达量则至少在两种非生物胁迫中显著下调,且NnCKX2、3、4、7和9正向响应逆境相关植物生长调节剂,推测NnCKX2、7和9在响应非生物胁迫中的作用尤其重要。本研究将为进一步开展NnCKXs的功能研究和逆境响应机制提供参考。
The cytokinin oxidase/dehydrogenase(CKX)enzyme,which catalyzes the irreversible degradation of endogenous cytokinins by cleaving their unsaturated side chains,plays a crucial role in plant stress responses.To explore its potential functions in lotus(Nelumbo nucifera),bioinformatics methods were employed to identify the CKX gene family members,and qRT-PCR technology was used to investigate the expression patterns of NnCKX genes under various tissues and abiotic stress conditions.The results reveal that 9 NnCKX genes are grouped into 4 subgroups,and their promoter regions contain numerous cis-acting elements responsive to abiotic stress and plant hormones.Furthermore,all NnCKXs have active binding sites capable of interacting with isopentenyl adenine(iP).NnCKXs are ubiquitously expressed across different tissues,exhibiting significant variability in their expression levels.Notably,they exhibit higher expression in roots and leaves.The NnCKX genes exhibit differential responses to low temperature,salt,and anaerobic stress,as well as induction by stress-related plant growth regulators(abscisic acid and methyl jasmonate)and growth-related plant growth regulators(auxin,gibberellin,and cytokinin).Specifically,genes like NnCKX2,3,5,6,and 9 show significant upregulation under at least two types of abiotic stress,whereas genes like NnCKX1,4,7 and 8 are significantly downregulated under similar conditions.Moreover,NnCKX2,3,4,7 and 9 genes positively respond to stress-related plant growth regulators,suggesting the potential importance of NnCKX2,7 and 9 in responding to abiotic stress.This study provides insights into the potential roles of NnCKX genes in lotus under stress conditions and serves as a reference for further functional studies and understanding stress response mechanisms involving NnCKX genes.
