麦冬不同部位红外光谱分析及成分含量的测定

摘要 试验旨在探究麦冬不同部位的红外光谱及多种成分含量差异,进行质量评价,为麦冬植株的综合开发利用提供科学依据。采用傅里叶变换红外光谱技术,对麦冬块根、须根、叶片、花、果实、花葶不同部位进行快速鉴别,采用硝酸铝显色法、Folin-Ciocalteu法、DNS法分别对麦冬不同部位的总黄酮、总酚、还原糖含量进行测定。结果表明:麦冬不同部位的一阶红外光谱显示,麦冬花、花葶、叶、块根、果实、须根存在较大的相似性,3300、2930、2110、1420、1030 cm^(-1)等位置存在明显的共有特征峰,经二阶导数光谱分析可进一步区分麦冬不同部位;麦冬不同部位均含有还原糖、总黄酮、总酚,但含量存在差异,其中果实中的还原糖、总酚、总黄酮含量均最高。 The experiment was aimed at exploring the infrared spectra of different parts of Ophiopogon japonicus and the differences in the contents of various substances,evaluating its quality,and providing a scientific basis for the comprehensive development and utilisation of the whole plant of Ophiopogon japonicus Fourier transform infrared(FTIR)spectroscopy was used for the rapid identification of different parts of Ophiopogon japonicus tuberous roots,fibrous roots,leaves,flowers,fruits and scapes,and aluminium nitrate chromatography,Folin-Ciocalteu method and DNS method were used for the determination of total flavonoids,total phenolics,and reducing sugars in different parts of Ophiopogon japonicus,respectively.The results showed that the first-order infrared spectra of the different parts of Ophiopogon japonicus showed that there were large similarities among the flowers,scapes,leaves,roots,fruits and fibrous roots of Ophiopogon japonicus,and there were obvious common characteristic peaks at the positions of 3300 cm^(-1),2930 cm^(-1),2110 cm^(-1),1420 cm^(-1),and 1030 cm^(-1),which could be further distinguished from the different parts of Ophiopogon japonicus by the second-order derivative spectral analysis.The different parts of Ophiopogon japonicus contain reducing sugars,total flavonoids and total phenols,but the content varies,among which the fruit of Ophiopogon japonicus has the highest content of reducing sugars,total phenols and total flavonoids.
