摘要
目的根据国际基于比吸收率(SAR,W/kg)的安全限值,研究职业暴露限值(0.4 W/kg)与公众暴露限值(0.08 W/kg)组合电磁暴露对雄性发育期大鼠生殖系统的影响。方法将72只健康清洁级雄性SD大鼠随机分为3组,分别为对照组(常规饲养)、假暴露组(饲养于微波暗室的单间内,但不进行辐照)和暴露组[于9:00—17:00以(0.4±0.052)W/kg的全身平均SAR值进行暴露,于17:00—次日9:00以(0.08±0.010)W/kg进行暴露],每组24只,持续暴露4周。于暴露后第0、7、14和30天,测定大鼠血清睾酮的浓度,睾丸组织内SOD活力与MDA含量以及附睾内精子质量。结果与对照组相比较,假暴露组各项指标无显著改变,差异无统计学意义(P>0.05)。与同期假暴露组相比较,暴露结束后第0、7天时暴露组大鼠睾丸组织SOD活力下降,MDA含量上升,差异均有统计学意义(P<0.05);暴露结束后第14、30天时暴露组大鼠睾丸组织内的SOD与MDA水平以及电磁暴露后暴露组大鼠各恢复时间点的血清睾酮水平均无明显改变。与同期假暴露组相比较,暴露后第0天时暴露组大鼠的精子活力、精子存活率、精子密度均下降,而精子畸形率显著上升,差异有统计学意义(P<0.05);暴露后第7、14天时暴露组精子密度均有所恢复,差异有统计学意义(P<0.05),而其余指标已无显著改变;暴露后第30天时暴露组的精子质量各项指标均无显著改变。结论在本实验条件下,职业与公众暴露限值组合电磁暴露可造成大鼠睾丸组织损伤,精子质量下降,而这些损伤在暴露后的30 d内可逐渐修复。
Objective Referring to the International Commission on Non-Ionizing Radiation Protection(ICNIRP)guideline(SAR,W/kg),to study the effects of occupational and public electromagnetic exposure levels in alternation on reproductive system in rats.Methods Totally 72 healthy clean-grade male SD rats were randomly divided into 3 groups,24 in each group,the control group(kept outside the anechoic chamber),the sham group(kept in anechoic chamber without irradiation)and the exposure group(exposure from 9:00 to 17:00 at(0.4±0.052)W/kg of whole body average SAR value,and from 17:00 to 9:00 the next day at(0.08±0.010)W/kg),and all experiment group lasted four weeks.On the 0th,7th,14th and 30th day after exposure,the serum testosterone concentration,SOD activity,MDA content in the testis tissue,and sperm quality in the epididymis were determined.Results Compared with the sham group,there was no significant change in the indicators of the control group(P>0.05).Compared with the sham group during the same period,the SOD activity of the testis tissue of the exposed group decreased on the 0th and 7th day after exposure,and the MDA content significantly increased(P<0.05).However,the levels of SOD and MDA in the testes of the exposed group on the 14th and 30th day after exposure,and the serum testosterone levels at each recovery time point after electromagnetic exposure did not change significantly(P>0.05).Compared with the sham group during the same period,the sperm motility,sperm survival rate,and sperm density of the rats in the exposure group decreased on 0th day after exposure,while the rate of sperm deformity increased significantly(P<0.05);The sperm density of the exposure group recovered on the 7th and 14th day,and the difference was statistically significant(P<0.05),while the other indicators had no significant changes;There was no significant change in all indexes of sperm quality in the exposure group on 30th day after exposure(P>0.05).Conclusion In the present study,SD male rats who were exposed to the occupational and public electromagnetic exposure levels in alternation for 28 consecutive days showed an oxidative damage in rat testicular and decrease of sperm quality,but these impacts can be self-recovered within 30 days.
