四君子汤合三子养亲汤对慢性阻塞性肺疾病小鼠肺功能及肺组织损伤的影响 被引量:11

Effects of Sijunzi Decoction(四君子汤)Combined with Sanzi Yangqin Decoction(三子养亲汤)on Lung Function and Lung Tissue Injury in Mice with COPD

摘要 目的 探讨四君子汤合三子养亲汤对慢性阻塞性肺疾病(chronic obstructive pulmonary disease, COPD)小鼠肺功能、肺组织的保护作用。方法 将40只小鼠按照随机数字表法平均分为空白组、模型组、西药组(盐酸氨溴索)、中药组(四君子汤合三子养亲汤),每组10只,采用烟熏联合脂多糖的方法建立慢阻肺小鼠模型。观察小鼠一般情况,动物肺功能仪检测肺功能,ELISA检测肺泡灌洗液(BALF)中IL-4、IL-8、IL-13的表达,HE染色观察肺组织病理。结果 与空白组比较,模型组0.15 s用力呼气容积与用力肺活量的比值(FEV_(0.15)/FVC)、每分钟最大通气量(MVV)明显下降;与模型组比较,中药组和西药组FEV0.15/FVC、MVV显著升高。与空白组比较,模型组最大呼气峰流速(PEF)显著下降;与模型组比较,中药组PEF显著升高,西药组PEF升高不明显,差异有统计学意义(P<0.05)。与空白组比较,模型组BALF中IL-4、IL-8、IL-13的表达增加,肺组织出现病理学损伤;与模型组比较,中药组和西药组BALF中IL-4、IL-8、IL-13的表达减少,肺组织损伤程度减轻。结论 四君子汤合三子养亲汤可下调小鼠肺部炎症因子水平,减轻肺组织结构的破坏,提高慢阻肺小鼠的肺功能。 Objective To explore the protective effects of Sijunzi Decoction(四君子汤)combined with Sanzi Yangqin Decoction(三子养亲汤)on lung function and tissue of mice with chronic obstructive pulmonary disease(COPD).Methods According to the random principle,forty mice were divided into blank group,model group,western medicine group(ambroxol hydrochloride)and Chinese medicine group(Sijunzi Decoction combined with Sanzi Yangqin Decoction),10 mice in each group.The COPD mouse model was established by the method of smoking combined with lipopolysaccharide(LPS).The general conditions of the mice were observed.The lung function was detected by animal spirometer,and the expressions of interleukin-4(IL-4),interleukin-8(IL-8)and interleukin-13(IL-13)in bronchoalveolar lavage fluid(BALF)were detected by ELISA.HE staining was used to observe the pathological changes of lung tissue.Results Compared with those of the blank group,the forced expiratory volume in 0.15 second/forced vital capacity(FEV_(0.15)/FVC)and the maximal ventilatory volume per minute(MVV)in the model group were significantly decreased.Compared with those of the model group,the FEV0.15/FVC and MVV values in the Chinese medicine group and western medicine group were significantly increased.Compared with those of the blank group,the peak expiratory flow(PEF)of the model group was decreased significantly.Compared with that of the model group,the PEF of the Chinese medicine group was significantly increased,while that of the western medicine group was not significantly increased,and the difference was not statistically significant(P<0.05).Compared with those of the blank group,the expressions of IL-4,IL-8 and IL-13 in BALF of the model group were increased,and the lung tissue was subjected to pathological injury.Compared with those of the model group,the expressions of IL-4,IL-8 and IL-13 in BALF of the Chinese medicine group and western medicine group were decreased,and the degree of lung tissue damage was alleviated.Conclusion Sijunzi Decoction combined with Sanzi Yangqin Decoction can down-regulate the levels of pulmonary inflammatory factors in mice,reduce the damage of lung tissue structure and improve the lung function of COPD mice.
