基于数据挖掘与网络药理学探讨中医药治疗鼻炎用药规律和作用机制 被引量:1

Exploration of the Medication Rule and Mechanism of Traditional Chinese Medicine in Treating Rhinitis Based on Data Mining and Network Pharmacology

摘要 目的:基于数据挖掘与网络药理学研究鼻炎患者中药处方用药规律以及核心药物的作用机制。方法:收集武汉市第六医院2023年1-12月中医门诊就诊的鼻炎患者中药处方,运用Microsoft Office Excel、SPSS 25.0、Apriori算法对中药使用频次、使用剂量、性味归经、功效分类以及关联规则、系统聚类进行分析,以选择出核心中药。使用分析平台(TCMSP)及UniProt数据库进行核心药物靶点预测;通过DrugBank、GeneCards数据库进行鼻炎靶点预测;运用Venny 2.1.0平台得出交集靶点;运用软件Cytoscape 3.7.0构建“药物-成分-靶点”网络,并对核心中药的主要活性成分进行预测;通过STRING数据库构建蛋白相互作用(PPI)网络图并筛选出关键靶点:运用Metascape数据库,对交集靶点基因进行基因本体(GO)和京都基因与基因组百科全书(KEGG)富集分析。结果:共收集处方222首,包含279味中药,应用频率前3位为白芷(158次),辛夷(145次),炒苍耳子(134次);药性以温、寒、平为主;药味以甘、辛、苦为主;中药饮片主要归经为肺、脾、胃、肝;多为解表药、补虚药、清热药。综合关联分析以及用药频次得出治疗鼻炎核心药物为炒苍耳子、白芷、辛夷、防风、茯苓、黄芪、陈皮、麸炒白术、薄荷。聚类分析得出5个聚类处方。网络药理学分析显示,核心药物靶点基因为254个,鼻炎靶点基因预测为871个,核心药物-鼻炎交集靶点78个,核心药物有效活性成分为槲皮素、辛夷脂素、β-谷甾醇等,核心中药治疗鼻炎关键靶点为TNF、IL-6、AKT1、IL-1β、PTGS2,GO功能分析得到1362个生物过程、50个细胞组分、106个分子功能,KEGG分析得到171条信号通路。结论:中药治疗鼻炎多为解表药、补虚药、清热药,用药特点为中医药治疗鼻炎患者提供思路。治疗鼻炎核心中药为炒苍耳子、白芷、辛夷、防风、茯苓、黄芪、陈皮、麸炒白术、薄荷。预测出核心中药通过多成分、多通路、多靶点来治疗鼻炎。 Objective:To investigate the medication patterns and molecular mechanisms of core drugs in traditional Chinese medicine prescriptions for patients with rhinitis based on data mining and network pharmacology.Methods:Traditional Chinese medicine prescriptions for rhinitis patients treated in the outpatient department of The Sixth Hospital of Wuhan from January 2023 to December 2023 were collected.Utilizing Microsoft Office Excel,SPSS 25.0,and the Apriori algorithm,the frequency of use,dosage,nature,meridian tropism,efficacy classification,and association rules of the Chinese herbs were analyzed through systematic clustering to select core Chinese medicines.The target prediction of core drugs was conducted using the analysis platform(TCMSP)and the UniProt database.Rhinitis target prediction was performed using the DrugBank and GeneCards databases.The intersection of target genes was obtained using the Venny 2.1.0 online platform.The“drug-component-target”network was constructed using the software Cytoscape 3.7.0,and the main active components of the core Chinese medicines were predicted.A protein-protein interaction(PPI)network was constructed using the STRING database,and key targets were screened.The Metascape online database was used to perform Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis on the intersection of target genes.Results::222 prescriptions were collected,including 279 types of Chinese herbs.The top three in terms of application frequency were Angelica dahurica(158 times),Magnolia liliflora(145 times),and fried Xanthium sibiricum(134 times).The nature of the drugs was mainly warm,cold,and neutral;the tastes were mainly sweet,pungent,and bitter;the Chinese herbal slices were mainly attributed to the lung,spleen,stomach,and liver meridians;and most were categorized as antipyretics,tonifying,and heat-clearing drugs.The core Chinese medicines for treating rhinitis,as determined by association analysis and frequency of use,were fried Xanthium sibiricum,Angelica dahurica,Magnolia liliflora,Saposhnikovia divaricata,Poria cocos,Astragalus membranaceus,Citrus reticulata,Atractylodes macrocephala,Mentha haplocalyx.Cluster analysis yielded five clusters of prescriptions.Network pharmacology analysis revealed that the core drug target genes were 254,predicted rhinitis target genes were 871,and the intersection of core drug-rhinitis target genes was 78.The effective active components of the core Chinese medicines were quercetin,magnolol,and beta-sitosterol,etc.The key targets for the treatment of rhinitis by core Chinese medicines were TNF,IL6,AKT1,IL1B,PTGS2.GO functional analysis yielded 1362 biological processes,50 cellular components,and 106 molecular functions;KEGG analysis identified 171 signaling pathways.Conclusion:The treatment of rhinitis with Chinese medicine mainly involves antipyretics,tonifying,and heat-clearing drugs.These characteristics provide insights into the treatment of rhinitis patients with traditional Chinese medicine.The core Chinese medicines for treating rhinitis are fried Xanthium sibiricum,Angelica dahurica,Magnolia liliflora,Saposhnikovia divaricata,Poria cocos,Astragalus membranaceus,Citrus reticulata,Atractylodes macrocephala,Mentha haplocalyx.The predicted core Chinese medicines treat rhinitis through multiple components,multiple pathways,and multiple targets.
