摘要
富含半胱氨酸的S100A3是S100蛋白家族成员,其表达具有组织和细胞特异性。本研究利用基因克隆技术将小鼠S100A3克隆到原核表达载体p ET28a(+)上,经过原核表达纯化获得了高纯度的目的蛋白,制备了高效价的兔多克隆抗体。重组小鼠S100A3蛋白原核表达和纯化方法的建立及多克隆抗体的制备为其生物学功能研究提供了材料,并对其它S100蛋白家族的表达纯化具有一定的参考意义。
Cysteine-rich S100A3 is a member of S100 protein family. S100A3 shows a tissue- and cell-specific expression pattern. We constructed the mouse S100A3 expression vector, obtained S100A3 protein with high purity through expression and purification, and prepared high titer polyclonal antibody against S100A3. This strategy could facilitate the investigation of the biological function and have reference values for the expression and purification of other S100 proteins.
