摘要
目的 观察羧甲基茯苓多糖 (CMP)体内外给药对小鼠免疫功能的影响。方法 体外方法为将指数生长小鼠淋巴细胞或巨噬细胞与不同浓度的羧甲基茯苓多糖共培养 ,观察小鼠脾淋巴细胞的增殖作用和巨噬细胞吞噬中性红的能力。体内方法为每日给正常小鼠分别静脉注射不同剂量的羧甲基茯苓多糖 ,连续注射 1 0d ,用与体外同样方法观察CMP对小鼠脾淋巴细胞的增殖作用及对小鼠腹腔巨噬细胞吞噬功能的影响。结果 CMP静脉注射剂量在 5 ,1 0 ,50mg·kg- 1 时对小鼠脾淋巴细胞增殖、腹腔巨噬细胞吞噬功能均有明显的促进作用 ,体外给药浓度在 0 .1~ 50 μg·mL- 1 时对小鼠脾淋巴细胞增殖有直接的促进作用。对小鼠混合淋巴细胞反应也有明显的促进作用 ,CMP还显著增强ConA及LPS活化的小鼠脾淋巴细胞的增殖反应及小鼠腹腔巨噬细胞吞噬中性红的作用。
OBJECTIVE: To observe the immunopotentiating effect of carboxymethylpachymaran (CMP). METHODS: The effect of CMP on lymphocyte proliferation and phagositosis of peritoneal macrophages on neutral red were studied in mice. The lymphocyte proliferation with or without mitogen was assessed by MTT method in vitro. Phagositosis of peritoneal macrophages on neutral red was used to assess Phagositosis of peritoneal macrophages in vitr. RESULTS: CMP (5,10,50 mg·kg-1, ig for 10 days in vivo, 0.1 μg·mL-1, 0.1-50 μg·mL-1 in vitro) strengthed lymphocyte proliferation induced by ConA or LPS, promoted phagositosis of peritoneal macrophages on neutral red. Besides, CMP directly influenced mouse proliferation of spleen lymphocyte and stimulated mixed lymphocyte reaction (MLR) in vitro with dosage 0.1-50 μg·mL-1. CONCLUSION: CMP stimulated the immunological function in mice in different ways.
