摘要
目的探讨单纯疱疹病毒胸苷激酶(HSV-tk)基因转染联合抗病毒药物更昔洛韦(GCV)对人肺腺癌细胞系的杀伤作用。方法应用基因工程技术构建了携带单纯疱疹病毒胸苷激酶(HSV-tk)基因的逆转录病毒重组体pLXSN-tk,通过脂质体法转染PA317细胞,G418筛选出产重组病毒颗粒的生产细胞PA317-tk细胞。采用四甲基偶氮唑蓝(MTT)比色法检测HSV-tk/GCV系统对人肺腺癌细胞系GLC-82的生长抑制率。结果 重组逆转录病毒载体能有效地将HSV-tk(基因导入GLC-82细胞系内并使其获得对GCV的敏感性。结论肺腺癌细胞转染HSV-tk基因后,能有效地被GCV杀死。
Objective To evaluate the efficacy of gene therapy with herpes simplex virus thymidine kinase (HSV - tk) gene transfer followed by ganciclovir (GCV) therapy in lung adenocarcinoma.Methods A retrovirus expression vector pLXSN - tk was constructed with HSV- tk cDNA and a retroviral vector pLXSN. The pLXSN - tk vector was transferred into PA317 packaging cells by method of Liposome - mediated transfer. The retroviral vector - producing cells PA317 - tk was isolated in 500/ug/ml G418 medium. The cytotoxicity efficacy of HSV - tk/GCV system to lung adenocarcinoma cell line was evaluated by MTT assay. Results The recom-binant retroviral vector pLXSN - tk transfered HSV - tk gene into the genome of GLC - 82 cell line and made it sensitive to GCV. Conclusion The lung adenocarcinoma cell line transferred by HSV - tk gene can regress rapidly in response to GCV therapy.
