摘要
植物细胞培养可以补充野生肉苁蓉资源的不足。研究了培养条件对盐生肉苁蓉愈伤组织生长的影响。利用高效液相色谱分析系统比较了愈伤组织与天然盐生肉苁蓉主要成分的异同。结果显示:用改良Miller或改良MS培养基、25℃暗培养、激素配比为不含BA,IAA1mg/L,或配比为BA0.5mg/L,IAA3.0mg/L最有利于愈伤组织的生长。愈伤组织的主要成分指纹图谱与天然盐生肉苁蓉基本一致,只是愈伤组织的主要成分含量高于天然盐生肉苁蓉。实验表明:盐生肉苁蓉的愈伤组织培养可以生产与天然盐生肉苁蓉相同的活性成分。
Cistanche cell culture can be used to effectively supplement natural cistanche. This paper analyzes the effects of culture conditions on cistanche callus growth preceding cistanche cell cultures. The callus contents were compared with those of natural plants using high performance liquid chromatography (HPLC). The optimized conditions for cistanche callus growth in modified medium MS or Miller were 25 ℃ in darkness with mass concentrations of no BA and 1.0 mg/L IAA, or of 0.5 mg/L BA and 3.0 mg/L IAA. The 335 nm chromatograph of the callus is very similar to that of the natural plant. The concentrations of the main component in the cultured callus are more than that in the natural product, so the active substances can be effectively produced in the callus culture to replace the natural cistanche.
