摘要
目的 研究 β -胡萝卜素拮抗三氧化二镍的细胞毒作用。 方法 采用体外细胞培养法测定在β-胡萝卜素(2 5 ,5 0 ,10 0 μmol/L)作用下 ,体外染镍肺泡巨噬细胞 (AM)的存活率及活力。同时观察细胞内活性氧的产生 ,以及过氧化氢酶 (CAT)和谷胱甘肽过氧化物酶 (GSH Px)活力的变化。结果 在体外染镍肺泡巨噬细胞中加入不同浓度的 β -胡萝卜素后可提高该细胞的存活率和活力 ,可增强细胞内CAT、GSH Px的活性 ,其中以 10 0 μmol/L的 β -胡萝卜素最为明显。 结论 镍可致细胞脂质过氧化 ,β -胡萝卜素具有拮抗镍对肺泡巨噬细胞的细胞毒作用 ,可能与抗氧化损伤有关。
Objective To study the antagonism of β\|carotene on cytotoxic effect by Ni 2O 3.Methods Ni 2O 3 exposed alveolar macrophages were cultured in vitro with 25,50,and 100 μmol/L β\|carotene added respectively and their survival rates and activities were measured.In addition,the production of active oxygen,activity changes of catalase(CAT)and glutathione peroxidase(GSH Px)were observed.Results It revealed that β\|carotene elevated survival rates and activities of Ni 2O 3 exposed alveolar macrophages and enhanced the activities of CAT and GSH Px,especially when β\|carotene at 100 μmol/L .Conclusions β\|carotene could antagonize lipid peroxidation of alveolar macrophage induced by Ni 2O 3,which is probably due to its antioxidant property.
