摘要
对丙酮萃取分光光度法测定浮游植物体内叶绿素a的方法进行改进。改进法在样品经过冷冻、50℃热丙酮快速提取并放置2h后进行测定。对改进前后方法测定结果的方差分析表明,方法改进前后测定结果不存在显著性差异。方法改进后简化了操作,提取时间由原来的8～20 h缩短为1.5～2.5h。
The method for the determination of the concentration of phytoplankton chlorophyll a (the cool acetone method which used 90% cool acetone as the extraction agent of chlorophyll a) was improved. In the modified method,the sample was made to pass through freezing,quick extracting with hot acetone at 50℃,then determined after 2 hours' placement. The analysis result of variance showed that the two determining methods didn't have significant difference, but the modified method reduced the analytical course and the extracting time was shortened from 8-20 h to l.5-2.5 h.
