家蚕微孢子虫膜蛋白NbYOP1的鉴定和功能研究

摘要 YOP1蛋白在真核生物中是一个功能保守的蛋白质,是形成内质网管状结构的必需蛋白。YOP1蛋白整合插入到内质网膜上,通过诱导膜曲度来稳定管状内质网结构,其缺失会使内质网产生严重的畸变。对家蚕微孢子虫(Nosema bombycis,Nb)的Nbyop 1基因进行克隆和鉴定分析,结果表明,Nbyop 1 ORF全长516 bp,编码蛋白NbYOP1由171个氨基酸残基组成。NbYOP1的分子质量为19.5 kD,包含2个跨膜区。亚细胞定位预测NbYOP1定位于内质网,间接免疫荧光分析表明,在成熟微孢子虫细胞中NbYOP1定位于细胞质,部分NbYOP1蛋白呈点状聚集于细胞膜的边缘,亚细胞定位模式与内质网定位的预测一致。在成熟孢子细胞和感染后的BmN细胞中,NbYOP1的蛋白表达水平较高,说明YOP1在家蚕微孢子虫的不同生活时期均起着重要作用。该研究为进一步探究膜蛋白NbYOP1在家蚕微孢子虫内质网结构和功能中所发挥的作用奠定了基础。 The YOP1(YIP one partner)protein is a functionally conserved protein in eukaryotes and is an essential protein for the formation of tubular endoplasmic reticulum(ER).The YOP1 protein integrates into the ER membrane to stabilize the tubular ER structure by inducing membrane curvature,and its absence may cause severe distortion of the ER.The cloning and identification of the Nbyop 1 gene of Nosema bombycis(Nb)showed that the Nbyop 1 ORF had a total length of 516 bp,and the NbYOP1 protein was composed of 171 amino acid residues.NbYOP1 had a molecular mass of 19.5 kD and contained two transmembrane regions.Subcellular localization prediction showed that NbYOP1 localized to the ER,and indirect immunofluorescence analysis showed that NbYOP1 localized in the cytoplasm in mature spore cells,and part of NbYOP1 protein gathered in dot-pattern near the edge of the plasma membrane.The subcellular localization pattern was consistent with the prediction of ER localization of NbYOP1.The NbYOP1 protein had a relatively high expression in mature spores and infected BmN cells,indicating that YOP1 played an important role in different life periods of Nb.This study lays a foundation for further exploration of the role of membrane protein NbYOP1 protein in the structure and function of the ER in Nb.
