根瘤菌HH103调节因子NolR对大豆结瘤固氮影响及转录组差异分析 被引量:2

Effects of rhizobium HH103 regulatory factor NolR on soybean nodule fixation and transcriptome difference analysis

摘要 [目的]探究费氏中华根瘤菌HH103中调节因子NolR对黑龙江大豆结瘤固氮的影响与共生机制。[方法]将根瘤菌HH103及其敲除突变体ΔnolR接种于9种黑龙江省主栽大豆和Willams82进行盆栽实验,并选取了接种HH103及突变体ΔnolR的黑科60大豆根部进行转录组测序。[结果]与不接菌的CK相比,接种HH103和ΔnolR在一定程度上都能促进结瘤,但是不同品种的结瘤效果不同。与接种HH103相比,ΔnolR使黑科60的根瘤数与固氮酶活性分别显著降低了46.47%、15.76%。通过对黑科60根系进行转录组测序发现,接种HH103与接种ΔnolR的根系存在661个差异基因,其中上调表达基因538个,下调表达基因123个。nolR基因影响了黑科60中代谢过程、细胞解剖实体、催化活性等过程的功能基因表达,除此之外,nolR基因的缺失也使差异表达基因富集于苯丙素生物合成、异黄酮生物合成等途径。[结论]nolR的缺失会抑制黑科60的结瘤,这可能是通过调控植物与结瘤因子相关信号合成途径来影响黑科60的固氮结瘤,该研究为后续深入探究nolR基因对根瘤菌-大豆共生机制的影响提供了理论依据。 [Objective]To investigate the effect of NolR in Sinorhizobium fredii HH103 on nodule fixation in Heilongjiang soy-bean and its symbiotic mechanism.[Method]Rhizobium HH103 and its knockout mutantΔnolR were inoculated on 9 main soy-beans planted in Heilongjiang Province and Willams82 for pot experiment.The roots of soybean Heike 60 inoculated with HH103 and the mutantΔnolR were selected for transcriptome sequencing.[Result]Compared with CK without bacteria,HH103 andΔnolR can promote nodulation to some extent,but the nodulation effect of different varieties is different.Compared with inoculation with HH103,ΔnolR significantly reduced the nodule number and nitrogenase activity of Heike 60 by 46.47%and 15.76%,respectively.Transcriptome sequencing of the roots of Heike 60 showed that there were 661 differentially ex-pressed genes between the roots inoculated with HH103 and those inoculated withΔnolR,including 538 up-regulated genes and 123 down-regulated genes.Gene nolR affects the expression of functional genes in the metabolic process,cellular anatomi-cal entity,catalytic activity and other processes in Heike 60.In addition,the absence of nolR gene also led to the enrichment of differentially expressed genes in phenylpropanoid biosynthesis,isoflavone biosynthesis.[Conclusion]The absence of nolR can inhibit the nodulation of Heike 60,which may affect the nitrogen-fixing nodulation of Heike 60 by regulating the signal synthe-sis pathway related to nodulation factors in plants.This study provides a theoretical basis for further investigation of the influ-ence of nolR gene on the mechanism of rhizobia-soybean symbiosis.
