黄龙舌草颗粒薄层色谱鉴别与含量测定 被引量:1

Identification and content determination of Huanglong Shecao granules by thin-layer chromatography

摘要 目的建立黄龙舌草颗粒的薄层色谱鉴别及黄芩的含量测定方法以控制质量。方法采用薄层色谱(TLC)对赤芍、龙胆进行定性鉴别;采用高效液相色谱法(HPLC)测定黄芩苷的含量,流动相:甲醇–0.2%磷酸溶液(47∶53);色谱柱:Kromasil C18(4.6 mm×250 mm,5μm);流速:1.0 ml/min;柱温:30℃;检测波长:280 nm。结果赤芍、龙胆的薄层色谱鉴别方法专属性强,其他成分无干扰;黄芩苷浓度在18.44~73.76μg/ml范围内与峰面积呈良好的线性关系(r=0.9999);平均回收率为100.05%,RSD=0.17%;精密度试验RSD=0.22%;稳定性试验表明含量测定溶液8 h内稳定;重现性试验RSD=0.96%,方法重现性良好。结论该方法简便、准确、重复性好,可用于黄龙舌草颗粒的质量控制。 Objective To establish a thin-layer chromatography identification method for Huanglong Shecao granules and a content determination method for Scutellaria baicalensis to control the quality.Methods Thin-layer chromatography(TLC)was used for qualitative identification of red peony and gentian.The content of baicalin was determined by high-performance liquid chromatography(HPLC),with a mobile phase of methanol 0.2%phosphoric acid solution(47∶53),chromatographic column:Kromasil C18(4.6 mm×250 mm,5μm),flow rate:1.0 ml/min,column temperature:30℃,detection wavelength:280 nm.Results The thin-layer chromatography identification method for red peony and gentian had strong specificity,and other components had no interference.There was a good linear relationship between baicalin concentration and peak area in the range of 18.44-73.76μg/ml(r=0.9999).The average recovery rate was 100.05%,RSD=0.17%,precision test RSD=0.22%.The stability test showed that the content determination solution was stable within 8 hours.The reproducibility test RSD was 0.96%,and the method had good reproducibility.Conclusion This method is simple,accurate,and has good repeatability,and can be used for quality control of Huanglong Shecao granules.
