摘要
目的探讨肺心宁汤对肺心病大鼠血栓素A2(TXA2)、前列环素(PGI2)、环氧化酶(COX)-2的表达和管壁面积百分比(WA%)、管腔面积百分比(LA%)的影响。方法从50只雄性SD大鼠中随机抽取10只作为空白组;其余大鼠通过浓度为1%的野百合碱(MCT)溶液,按照60 mg/kg腹腔注射1次,14 d后即形成肺心病大鼠模型,随机分为模型组、中药组、西药组、中药+西药组。中药组、西药组、中药+西药组连续给药14 d。治疗14 d后检测TXA2、PGI2、COX-2的表达以及WA%、LA%。结果与空白组大鼠比较,模型组大鼠PGI2表达和LA%降低,TXA2、COX-2表达和WA%升高(P<0.05)。与模型组大鼠比较,中药组、西药组、中药+西药组PGI2表达升高,TXA2、COX-2表达降低(P<0.05);WA%和LA%出现了不同程度的改善趋势,但差异无统计学意义(P>0.05)。结论肺心宁汤可以降低肺心病大鼠TXA2、COX-2的表达,增加PGI2的表达;对于大鼠WA%和LA%虽有不同程度的改善,但差异无统计学意义。
Objective To observe the effects of Feixinning decoction on the expression of PXA2,PGI2,COX-2 together with WA%and LA%.Methods Ten of fifty male SD rats were randomly selected as the blank group,and the rest rats were injected with monocrota⁃line(MCT)solution with a concentration of 1%at a dose of 60 mg/kg intraperitoneally for one time,and rat model with pulmonary heart disease was established 14 days later.After successful modeling,the rats were randomly divided into model group,the Chinese medicine group,the Western medicine group,and the Chinese and Western medicine group.The expressions of PXA2,PGI2 and COX-2 together with WA%and LA%were detected after 14 days of continuous administration in each group.Results Compared with rats in the blank group,the expression of PGI2 and LA%decreased,while the expression of TXA2,COX-2 and WA%increased in the model group(P<0.05).Compared with the model rats,the expressions of PGI2 were increased and the expressions of TXA2 and COX2 were decreased in the three treatment groups.Compared with the model group,the WA%and LA%showed an improvement trend in different degrees,but the differences were not statistically significant(P>0.05).Conclusion Feixinning decoction can improve the ex⁃pression of PGI2 in rats with pulmonary heart disease,increase the expression of TXA2 and COX-2.For WA%and LA%,although there is no statistical difference,all of them have improved to varying degrees.
