摘要
为分析陕西省牛病毒性腹泻病毒(BVDV)分子变异情况,采集陕西省某牛场腹泻患牛粪便样品,经处理后接种至MDBK细胞后,进行病毒分离鉴定并对分离的BVDV进行E2基因的克隆和遗传进化分析。结果显示,样品经过RT-PCR鉴定后接毒,第4天出现明显的细胞病变(CPE),通过设计特异性引物对收获的病毒成功扩增出BVDV E2基因,证明分离的病毒为BVDV。对该病毒的E2基因进行序列分析,所得序列与GenBank上已发表的BVDV毒株的核苷酸序列同源性为84.5%~100%,氨基酸序列同源性为82.1%~100%。结果表明,成功分离到牛病毒性腹泻病毒并进行了E2基因的克隆与序列分析,研究结果可为陕西地区BVDV的流行病学、致病性及生物学特性研究提供依据,为BVDV的区域流行及有效防控提供参考,为新疫苗的研发提供地方种毒。
In order to study the molecular variation of bovine viral diarrhea virus(BVDV)in Shaanxi province,fecal samples of diarrhea-affected cattle from a cattle farm in Shaanxi province were collected,treated and then inoculated into MDBK cells,then virus isolation and identification were carried out,and the isolated BVDV was subjected to the cloning of the gene for E2 and genetic evolution analysis.The results showed that obvious cytopathic effects(CPE)appeared on the fourth day of inoculation,and the BVDV E2 gene was successfully amplified by designing specific primers for the harvested virus,which proved that the isolated virus was BVDV.Sequence analysis was performed on the E2 gene of the virus,and the homology of the obtained sequence with the nucleotide sequences of the published strains of BVDV in GenBank ranged from 84.5%to 100%,and the amino acid homology ranged from 82.1%to 100%.In the present study,the bovine viral diarrhea virus was successfully isolated and the E2 gene was cloned and sequenced,which provides some references for the epidemiological investigation,prevention and control of BVDV in Shaanxi.
