摘要
膝骨关节炎是最普遍的关节疾病,其特征是膝关节软骨退化、滑膜炎症以及关节周围和软骨下骨的变化。最新研究报道了几种关键的信号通路,它们是膝骨关节炎病理状态下细胞和分子的关键调节因子和激活因子。而Wnt/β-catenin正是这样独特的信号通路,该特定通路中分子的激动剂和拮抗剂可作为膝骨关节炎治疗的潜在候选者。此通路应严格调节,以维持软骨在过度激活或抑制Wnt/β-catenin信号通路下的稳态。Wnt信号级联的复杂性及其药物对其机制的影响仍未完全了解,这迫使我们需要进一步地研究并开发有效的治疗方法来治疗膝骨关节炎。根据目前对转基因小鼠体内、体外研究以及临床观察等,该文对Wnt/β-catenin通路中的相关分子通过转导所产生的级联反应提供独特见解;并且对Wnt/β-catenin信号通路是如何影响膝骨关节炎中的软骨细胞和滑膜组织,以及病理状态下中医与西医的治疗方式是如何靶向该途径来改善临床症状进行归纳与总结。
Knee osteoarthritis is the most common joint disease and is characterized by degeneration of the knee cartilage,synovial inflammation,and changes in the periarticular and subchondral bones.Recent studies have reported several key signaling pathways that are key regulatory and activators of cells and molecules in the pathological state of knee osteoarthritis.Wnt/β-catenin is a unique signaling pathway,and agonists and antagonists of molecules in this specific pathway can be potential candidates for the treatment of knee osteoarthritis.This pathway should be strictly regulated to maintain cartilage homeostasis under overactivation or inhibition of the Wnt/β-catenin signaling pathway.The complexity of the Wnt signaling cascade and the effects of drugs on its mechanisms are still not fully understood,forcing the need for further research and the development of effective therapies to treat knee osteoarthritis.Based on current in vivo and in vitro studies and clinical observations of transgenic mice,this paper provides additional insights into the cascade reactions of related molecules in the Wnt/β-catenin pathway through conduction.In addition,how the Wnt/β-catenin signaling pathway affects chondrocytes and synovial tissue in knee osteoarthritis and how the treatment methods of traditional Chinese medicine and Western medicine in pathological state target this pathway to improve clinical symptoms were summarized.
