摘要
目的:分离具有角蛋白酶和纤维素酶活性的菌株,并对获得的菌株进行鉴定。方法:从一处竹林散养鸡场地表取得土样,以羽毛粉培养基和羧甲基纤维素钠培养基作为筛选培养基,筛选同时对角蛋白和纤维素具有降解能力的菌株,对所获得的菌株进行16S rDNA序列分析、生理生化试验及酶活性测定。结果:分离得到1株菌株,编号为WYM41;对其16S rDNA序列进行分析显示,与苏云金芽孢杆菌的相似度为98.84%;经细菌形态学观察及生理生化特性鉴定初步确定其属于芽孢杆菌属;降解实验结果显示,菌株WYM41对羽毛、羧甲基纤维素钠和滤纸条具有较强的降解能力;酶活性测定结果显示,每100 mL发酵液中蛋白酶酶活力为(559.78±43.831) U。结论:菌株WYM41具有芽孢杆菌的典型特征,能够产生角蛋白酶和纤维素酶,因此在微生物资源的开发和利用上具有应用前景。
Objective:To isolate strains with keratinase and cellulase activities,and to identify the obtained strains.Methods:Soil samples were taken from the ground of a free-range chicken farm in a bamboo forest.The strains capable of degrading both keratin and cellulose were selected by using feather powder medium and sodium carboxymethyl cellulose medium,respectively.16S rDNA sequence analysis,physiological and biochemical tests,and enzyme activity measurements were carried out.Results:A strain was isolated,and numbered as WYM41.Its 16S rDNA sequence was compared,and the similarity with Bacillus thuringiensis was 98.84%.Through morphological observation of bacteria and identification of physiological and biochemical characteristics,the strain was preliminarily identified as Bacillus genus.The degradation experiment showed that strain WYM41 had strong degradation ability on feathers,carboxymethyl cellulose sodium,and filter paper.Enzyme activity measurement showed that the enzyme activity was(559.78±43.831) U per 100 mL fermentation broth.Conclusion:The strain WYM41 has typical characteristics of Bacillus and can produce keratinase and cellulase,which has a promising application in the development and utilization of microbial resources.
