药食同源组方对RAW 264.7巨噬细胞的免疫调节作用 被引量:1

Immunomodulatory Effects of Drug-Food Homologous Prescriptions on RAW 264.7 Macrophages

摘要 以红参、阿胶、茯苓、山药、枸杞为原料研制出3个药食同源组方并探究其对小鼠单核巨噬细胞(RAW264.7细胞)的免疫调节活性及作用机制。采用Cell Counting Kit-8 (CCK-8)法检测不同浓度制剂作用下的RAW264.7细胞增殖情况;利用酶联免疫(ELISA)法分析3个药食同源组方对RAW 264.7巨噬细胞吞噬能力、一氧化氮(NO)、活性氧(ROS)、白细胞介素-1β (IL-1β)、白细胞介素-6 (IL-6)和肿瘤坏死因子(TNF-α)细胞因子分泌量的影响。50~200μg/mL时, 3个药食同源组方对RAW 264.7巨噬细胞无毒害作用。ELISA试验结果表明,质量浓度200μg/mL时,药食同源组方可明显促进RAW 264.7巨噬细胞的产NO能力(P<0.05)。此外, 3个药食同源组方对RAW 264.7巨噬细胞的吞噬能力、细胞内ROS生成量及细胞因子分泌量均有不同程度的促进作用。采用网络药理学构建成分-疾病靶点交集基因的PPI网络图,结果表明3个药食同源组方能通过调控AKT1、STAT3、TNF、EGFR、SRC、ESR1、MMP9等靶点提高机体免疫力。研究表明, 3个药食同源组方均能通过提高免疫应答发挥免疫调节作用,具有多成分、多靶点、多通路的特点。 Three different formulations were prepared with red ginseng, Ejiao, Tuckaia, yam and wolfberry as RAW materials,and their immunomodulatory activity and mechanism on mouse mononuclear macrophages(RAW 264.7 cells) were investigated.Cell Counting Kit-8(CCK-8) was used to detect the proliferation of RAW 264.7 cells under different concentrations of preparations.The effects of drug-food homologous prescriptions on the phagocytic capacity, nitric oxide(NO), reactive oxygen species(ROS), interleukin-1β(IL-1β), interleukin-6(IL-6) and tumor necrosis factor(TNF-α) cytokine secretion were determined by enzyme-linked immunoassay(ELISA). At the concentration of 50-200 μg/mL, drug-food homologous prescriptions had no toxic effect on RAW 264.7 macrophages. The results of ELISA experiments showed that the preparation of 200 μg/mL could significantly promote RAW 264.7 macrophages' NO-producing capacity(P<0.05). In addition, the preparation promoted the phagocytosis ability, intracellular ROS production and cytokine secretion of RAW 264.7 macrophages to varying degrees.Using network pharmacology, a PPI network diagram of the intersection of ingredients and disease target genes was constructed.The results showed that drug-food homologous prescription could enhance immune function by regulating targets such as AKT1, STAT3, TNF, EGFR, SRC, ESR1 and MMP9. It demonstrated that the drug-food homologous prescriptions all exerted immunomodulatory effects by enhancing immune responses, and they exhibited characteristics of multiple components, multiple targets, and multiple pathways.
