摘要
目的探讨白细胞介素35(IL-35)、Beclin-1和乳酸脱氢酶(LDH)在乳腺癌患者外周血和组织中的表达情况及相关性。方法选取2022年4—9月在上海市嘉定区妇幼保健院诊治的原发性乳腺癌的女性患者共44例,对照组46例,ELISA测定外周血血浆中IL-35和Beclin-1水平;全自动生化分析仪上分析血清LDH水平;免疫组化分析EBI3、Beclin-1和LDHA蛋白抗体在组织中的表达水平。结果与对照组相比,外周血中IL-35[(19.30±3.63)pg/mL]和LDH[(191.50±43.07)U/L]水平高表达(P<0.05),Beclin-1[(2.39±0.517)ng/mL]低表达(P<0.05);相关性分析显示IL-35、LDH与Beclin-1分别都呈负相关,而IL-35与LDH之间呈正相关。ROC曲线分析显示,各项外周血指标联合检测诊断的AUC值为0.861,高于IL-35(0.768)、Beclin-1(0.733)及LDH(0.657)单一诊断AUC值(P<0.05)。EBI3特异性抗体间接表示的IL-35,LDHA间接表示的LDH以及Beclin-1蛋白抗体的免疫组化结果显示,与癌旁组织相比,乳腺癌组织中IL-35和LDH蛋白水平高表达,Beclin-1低表达。结论乳腺癌患者外周血和组织中IL-35和LDH高表达,Beclin-1低表达,IL-35可能与自噬水平呈负相关,与糖代谢水平呈正相关。
Objective To investigate the expression and correlation of IL-35,Beclin-1,and LDH in peripheral blood and tissues of breast cancer patients.Methods From April to September 2022,44 female patients with primary breast cancer who were diagnosed and treated in Shanghai Jiading Maternal and Child Health Hospital were selected,and 46 women in the control group were selected.The levels of IL-35 and Beclin-1 in peripheral blood plasma were measured by ELISA.LDH levels of serum were analyzed on a fully automated biochemical analyzer.The levels of EBI3,Beclin-1,and LDHA protein expressions in tissues were detected by Immunohistochemical.Results Compared with the control group,IL-35[(19.30±3.63)pg/mL]and LDH[(191.50±43.07)U/L]in peripheral blood were higher expressed(P<0.05),while Beclin-1[(2.39±0.517)ng/mL]levels were lower(P<0.05).Correlation analysis shows that IL-35,LDH,were both negatively correlated with Beclin-1,while IL-35 and LDH were positively correlated with each other.According to the ROC curve,the AUC value diagnosed by the combined detection of various peripheral blood indicators was 0.861,which was higher than the three single diagnoses(P<0.05).Immunohistochemical results of EBI3,Beclin-1,and LDHA,protein antibodies showed that compared with adjacent tissues,the levels of IL-35 and LDH protein in breast cancer tissues were higher,and the levels of Beclin-1 protein were lower.Conclusion In peripheral blood and tissues of breast cancer patients,IL-35 and LDH had a higher expression,while Beclin-1 had a lower expression.IL-35 may be negatively correlated with autophagy level,and positively correlated with glucose metabolism level.
