基于玉米醇溶蛋白/酪蛋白酸钠/壳聚糖构建紫苏醛递送体系及其抑菌效果探究

Construction of perillaldehyde delivery system based on zein/casein sodium/chitosan and its antifungal effects

摘要 目的构建荷载紫苏醛的递送体系,降低紫苏醛的刺激性,提高其亲水性应用。方法利用液-液分散法制备荷载紫苏醛的玉米醇溶蛋白(zein)纳米粒,加入稳定剂酪蛋白酸钠(casein sodium,NaCas),再结合壳聚糖(chitosan,CTS)制得递送体系。体系构建后对纳米粒的粒径、电位、包封率、冻干复溶性等进行表征评价。在此基础上,通过分析黄曲霉的菌丝生长、生物量、孢子萌发和次级代谢产物黄曲霉毒素B1(aflatoxin B1,AFB1)含量,以比较游离紫苏醛和荷载紫苏醛的zein-NaCas-CTS纳米粒的抑菌效果。结果当zein/NaCas/CTS添加量比为1.00:1.00:2.00时,荷载紫苏醛的zein-NaCas-CTS纳米粒粒径为342.27 nm,电位为+49.37 mV,包封率达到98.18%。荷载紫苏醛质量浓度为1/3 mg/mL的zein-NaCas-CTS纳米粒对菌丝生长、生物量和AFB1的抑制效果显著高于相同质量浓度的游离紫苏醛,而孢子萌发率无显著差异。结论基于zein-NaCas-CTS的递送体系可显著提升紫苏醛的物理特性和抑菌效果。 Objective To construct a delivery system loaded with perillaldehyde,reduce the irritation of perillaldehyde and improve its hydrophilic application.Methods Perillaldehyde-loaded zein-casein sodium(NaCas)nanoparticles were prepared by liquid-liquid dispersion method,along with chitosan(CTS)was added to stabilize NaCas.The particle size,potential,encapsulation rate and resolubility of the nanoparticles were evaluated after the system was constructed.Furthermore,the antifungal effects of free perillaldehyde and zein-NaCas-CTS nanoparticles loaded with perillaldehyde were compared by analyzing the mycelial growth,biomass,spore germination and secondary metabolite aflatoxin B1(AFB1)content of Aspergillus flavus.Results The particle size of zein-NaCas-CTS nanoparticles loaded with perillaldehyde was 342.27 nm,the potential was+49.37 mV,and the encapsulation rate reached 98.18%when the zein/NaCas/CTS mass ratio was 1.00:1.00:2.00.The inhibitory effect of zein-NaCas-CTS nanoparticles loaded with 1/3 mg/mL perillaldehyde on mycelial growth,biomass and AFB1 content was significantly higher than that of free perillaldehyde at the same concentration,while had no effects on the spore germination rate.Conclusion zein-NaCas-CTS system can improve the physical characteristics and antifungal effects of perillaldehyde.
