摘要
为了了解贵州省猪流行性腹泻(porcine epidemic diarrhea,PED)的流行情况,试验对猪流行性腹泻病毒(Porcine epidemic diarrhea virus,PEDV)贵州流行株(GZ2022)N基因序列进行克隆、测序,并利用DNAStar、ProtParam、ProtScale等生物信息学分析软件对PEDV-GZ2022株N基因的同源性和遗传进化地位,以及N蛋白的分子特征和潜在功能进行分析预测。结果表明:PEDV-GZ2022株N基因大小约为1364 bp,编码441个氨基酸;PEDV-GZ2022株N基因核苷酸序列与GenBank中的PEDV参考毒株N基因的相似性为95.5%~99.8%,与PEDV经典毒株CV777 N基因核苷酸序列的相似性为95.6%,与SDLY2020毒株N基因核苷酸序列的相似性高达99.8%,PEDV-GZ2022毒株与经典毒株CV777关系相隔较远,与PEDV变异株GⅡ-b位于同一分支;PEDV-GZ2022株N蛋白中含有较多的疏水性氨基酸,为疏水性蛋白;该蛋白质存在4个N糖基化位点和37个O糖基化位点,有31个丝氨酸、14个苏氨酸及3个酪氨酸可能被磷酸化,并且抗原指数较好。说明PEDV-GZ2022株为GⅡ-b变异株,PEDV-GZ2022 N蛋白可能具有良好的抗原性,可以作为潜在的诊断抗原。
In order to understand the epidemiology of porcine epidemic diarrhea(PED)in Guizhou Province,in the experiment,the full sequence of the N gene of the Guizhou epidemic strain(GZ2022)of Porcine epidemic diarrhea virus(PEDV)was cloned and sequenced;bioinformatics analysis software such as DNAStar,ProtParam and ProtScale were used to analyze and predict the homology and genetic evolutionary status of the N gene of the PEDV-GZ2022 strain,as well as the molecular characteristics and potential functions of the N protein.The results showed that the size of the N gene of PEDV-GZ2022 strain was about 1364 bp,encoding 441 amino acids;the similarity of the nucleotide sequence of the N gene of PEDV-GZ2022 strain to the N gene of the PEDV reference strains in GenBank was 95.5%-99.8%;its similarity to the nucleotide sequence of the N gene of the classical PEDV strain CV777 was 95.6%,and its similarity to the nucleotide sequence of the N gene of the SDLY2020 strain was as high as 99.8%;the PEDV-GZ2022 strain was distant from the classical strain CV777,and be longed to the same branch with the PEDV mutant strain GⅡ-b.The N protein of PEDV-GZ2022 strain contained more hydrophobic amino acids and was a hydrophobic protein;the protein existed 4 N glycosylation sites and 37 O-glycosylation sites,31 serines,14 threonines and 3 tyrosines might might be phosphorylated,and had a good antigenic index.The results suggested that the PEDV-GZ2022 strain was a GⅡ-b mutant and that the PEDV-GZ2022 N protein might have good antigenicity and could be used as a potential diagnostic antigen.
