青海草原毛虫幼虫感器类型及GqinOBPs表达分析 被引量:6

摘要 为明确青海草原毛虫(Gynaephora qinghaiensis)幼虫感受器的类型及气味结合蛋白GqinOBPs基因在嗅觉识别过程中的功能。本研究通过扫描电镜观察青海草原毛虫6龄幼虫的感受器种类和形状,并通过实时荧光定量检测技术测定GqinOBPs基因在卵、幼虫和蛹中的表达情况。结果显示,青海草原毛虫幼虫触角上分布有7种感器,分别是腔锥形感器(SCo)、4种栓锥形感器(SSTⅠ-Ⅳ)和2种刺形感器(SChⅠ-Ⅱ);头部及口器周围共分布9种感器,分别是3种锥形感器(SBⅠ-Ⅲ),2种刺形感器(SChⅠ-Ⅱ),3种栓锥形感器(SSTⅠ-Ⅲ)和1种毛型感器(ST);足上分布有5种感器,分别是4种刺形感器的(SChⅠ-Ⅳ)和1种锥形感器;毒腺上只有Bohm氏鬃毛感器。基因表达谱分析显示,青海草原毛虫15个OBPs基因在卵、幼虫和蛹中均有不同程度的表达,其中GqinOBP4和GqinOBP10表达量较高。本研究旨在为青海草原毛虫化学感受机制奠定基础,同时也为寻找青海草原毛虫绿色防治的分子靶标提供理论依据。 To clarify the type of receptor and the function of GqinOBPs gene in the process of olfactory recognition,we observed the receptor species and shapes of 6th instar larvae of G.qinghaiensis by SEM and determined the expression of GqinOBPs gene in eggs,larvae,and pupae by quantitative real-time detection technology in this study.The results showed that there were 7 kinds of sensilla on the antennae of G.qinghaiensis larvae,1 kind of sensilla coeloconica,4 kinds of sensilla styloconica(SSTⅠ-Ⅳ)and 2 kinds of sensilla chaetica(SCh I-Ⅱ).There were 9 kinds of sensilla distributed around the head and mouthparts,3 sensilla basiconica(SBⅠ-Ⅲ),2 sensilla chaetica(SChⅠ-Ⅱ),3 sensilla styloconica(SSTⅠ-Ⅲ)and 1 sensilla trichodea(ST).There were 5 kinds of sensilla distributed on the foot,4 kinds of sensilla chaetica(SCh I-Ⅳ)and sensilla basiconica.There were only B hm bristles on the poison gland.Gene expression profiling showed that The 15 OBPs genes were expressed in eggs,larvae and pupae,including OBP4 and OBP10.This study laid the foundation for a deeper understanding of the chemosensory mechanism in G.qinghaiensis,and provided a theoretical basis for finding the molecular targets of green prevention and control in G.qinghaiensis.
