摘要
目的 :探讨吗啡对神经系统细胞的影响及作用机理。方法 :应用基因克隆技术克隆大鼠增殖细胞核抗原 ( PCNA)的 c DNA基因片段 ,并应用逆转录 PCR技术研究了吗啡对大鼠胶质细胞瘤C6细胞 PCNA基因表达的影响。结果 :0 .1、1 .0 mg· L-1吗啡作用于大鼠 C6细胞 2 4 h后 ,PCNAm RNA相对含量明显下降 ,且剂量越高 PCNA m RNA相对含量越低。结论 :吗啡对正常生长的大鼠 C6细胞的增殖具有一定的抑制作用 ,其机理可能是通过抑制大鼠 C6细胞 PCNA的转录过程 ,使 PCNA转录水平降低 。
Objective:To study the effect of morphine on nerve system cells and its mechanism. Methods: Using gene cloning technique to get the recombinant of PCNA gene fragment; Using RT PCR method to study the effects of morphine on the level of PCNA gene transcript in C6 rat glioma cells treated with different doses of morphine.Results: The level of PCNA gene transcript in C6 rat glioma cells was down regulated after treated with 0.1,1.0 mg·L -1 morphine for 24 hours in a dose dependent way. Conclusion:Morphine has a certain inhibitory effect on the proliferation of cultured C6 cells. Its mechanism should be that morphine inhibits the transcript of PCNA, thereby inhibits the anabolism of nucleic acid.
