摘要
研究葛根提取物对去卵巢大鼠阴道、子宫及垂体 -性轴激素变化的影响。选用 6 0只雌性Wistar大鼠 ,体重 2 5 0～ 2 6 0g ,除正常组和假手术组外 ,其余各组动物摘除双侧卵巢。动物随机分为正常组 ,模型组 ,雌二醇组 ,假手术组 ,葛根提取物低剂量组 ,葛根提取物高剂量组。各组给药 6周后 ,麻醉后颈动脉取血分离血清 ,测血清中雌二醇 (E2 )、促黄体生成素 (LH)等激素 ,剥离阴道与子宫称重。HE染色观察子宫组织形态学变化。结果显示葛根提取物能显著提高去卵巢大鼠阴道和子宫重量 ;改善子宫萎缩状况 ;葛根提取物能增加去卵巢大鼠血清E2 ,含量 ,降低LH含量。说明葛根提取物增加去卵巢大鼠阴道、子宫重量和改善萎缩状况并改变其血中激素水平 。
The effects of the extract of gegen (the dried Root of Pueraria lobata or Pueraria thomsonii) on the vagina and uterus and on hormones in the pituitary sex gland axis were investigated in the ovariectomized rats. 60 Female Wistar rats with the body weight between 250 g to 260 g were randomly divided into 6 groups, namely the normal control group, model group, estradiol group, sham operation group, small dose of gegen extract group and large dose of gegen extract group. The rats in all groups except the normal control group and sham operation group, were bilaterally ovariectomized. After the administration of gegen extract for 6 weeks, the rats in each group were anesthetized for collecting carotid blood. And then, the levels of hormones such as E 2 and LH in the serum of the carotid blood were detected and the weight of the isolated vagina and uterus was measured respectively. The histomorphological changes of the uterus were observed by the use of HE staining. The results showed that gegen extract could markedly increase the weight of the vagina and uterus, relieve uterine atrophy, raise the level of E 2, and lower the level of LH. The results of the study can provide experimental evidence for developing new drugs for treating climacteric.
