基于炎症与糖脂代谢探讨连陈汤对肥胖小鼠体质量的影响 被引量:4

Impacts of Lianchen Tang on Body Mass in Obesity Mice Based on Inflammation and Glycolipid Metabolism

摘要 目的:探讨连陈汤对肥胖小鼠体质量、炎症因子以及糖脂代谢水平的影响。方法:选取C57BL/6J健康雄性小鼠40只,随机分为4组各10只。空白组予普通饲料,模型组、辛伐他汀组、连陈汤组予高脂饲料,建立肥胖小鼠模型。造模成功后空白组普通饲料喂养、纯净水灌胃;模型组高脂饲料喂养、纯净水灌胃;辛伐他汀组高脂饲料喂养、辛伐他汀灌胃;连陈汤组高脂饲料喂养、连陈汤灌胃。每日1次,连续灌胃4周。检测糖脂代谢指标[甘油三酯(triglyceride,TG)、总胆固醇(total cholesterol,TC)、空腹血糖(fasting plasma glucose,FPG)、空腹胰岛素(fasting insulin,FINS)、胰岛素抵抗指数(homeostasis model of insulin resistance index,HOMA-IR)]、血清炎症因子[肿瘤坏死因子α(tumor necrosis factorα,TNF-α)、白细胞介素6(interleukin-6,IL-6)、白细胞介素1β(interleukin-1β,IL-1β)]水平。结果:高脂饲料喂养的3组小鼠体质量高于普通饲料喂养的空白组,超过空白组体质量的20%;高脂饲料喂养的3组小鼠附睾脂肪组织重于空白组(P<0.01),而且存在大量脂滴积累;与模型组比较,连陈汤组小鼠体质量、TG含量、胰岛素抵抗指数及TNF-α、IL-6、IL-1β水平下降(P<0.01)。连陈汤组较辛伐他汀组改善小鼠体质量、胰岛素水平及炎症因子水平更优(P<0.05)。结论:连陈汤可降低肥胖小鼠附睾脂肪组织积累与炎性反应,改善血脂代谢、胰岛素抵抗,起减轻体质量作用。 Objective:To discuss the influence of Lianchen Tang on body mass,inflammatory factor and glycolipid metabolism in obesity mice.Methods:Forty C57BL/6J healthy male mice were chosen and randomized into four groups with ten ones in each group.The blank group was given common fodder,the model group,simvastatin group and Lianchen Tang group accepted high-fat feed,to establish obesity mice models.After successfully modeling,the blank group was drenched with common feed and purified water;the model group with high-fat feed and purified water;simvastatin group with high-fat feed and simvastatin;Lianchen Tang group with high-fat feed and Lianchen Tang,once each day,administering for four consecutive weeks.To detect the levels of glycolipid metabolism indexes including TG,TC,FPG,FINS and HOMA-IR,serum inflammatory factors covering TNF-α,IL-6 and IL-1β,the weight and morphology of fatty tissue in epididymis.Results:The mice in three groups fed a high-fat diet had higher body mass than the ones in the blank group fed an ordinary diet,exceeding 20%of the body mass of the mice in the blank group;large amount of lipid droplets accumulated in the fatty tissue of epididymis in the mice of the three groups fed a high-fat diet,and the weight of epididymis in three groups higher than of the blank group(P<0.01);compared with the model group,body weight,the contents of TG,insulin resistance index,the levels of TNF-α,IL-6 and IL-1βreduced in Lianchen Tang group(P<0.01).The improvements of body mass,the levels of insulin and inflammatory factors of Lianchen Tang group were more obvious than these of simvastatin(P<0.05).Conclusion:Lianchen Tang could lower the accumulation of fatty tissue and inflammation in epididymis of obesity mice,improve blood lipid metabolism,and better insulin resistance,thereby achieving the goals of losing weight.
