软枣猕猴桃茎黄酮的分离纯化及其片剂制备 被引量:4

摘要 以软枣猕猴桃茎为材料,分离获得黄酮粗提液,利用AB-8大孔吸附树脂对黄酮粗提物进行纯化,筛选不同因素(上样液浓度、上样液流速、上样液pH值、水洗体积、洗脱液浓度、洗脱剂流速、洗脱剂体积)对AB-8大孔吸附树脂吸附与解析能力的影响,利用正交试验法,探索黄酮添加量和不同辅料的选择对片剂制备工艺影响,并对片剂进行质量测定。经试验得到最佳纯化条件为:上样液浓度0.63 mg·mL^(-1),上样液流速3 mL·min^(-1),上样液pH值为5.0,水洗体积40 mL,洗脱液(乙醇)浓度80%,洗脱液流速3 mL·min^(-1),洗脱液体积30 mL,通过该方案获得的黄酮类化合物纯度可达71.25%。利用单因素和正交试验优化片剂的最佳配方,结果表明:正交试验法发现影响片剂的因素依次为C>B>A(即羧甲基纤维素钠的添加量>淀粉的添加量>软枣猕猴桃茎黄酮),最佳处方为A2B1C1。因此片剂最佳配方中软枣猕猴桃茎黄酮添加量为38%,淀粉添加量38%,羧甲基纤维素钠的添加量8%,甘露醇添加量16%。制成的片剂外观完整光滑、色泽均匀无杂质,硬度适中,崩解时限和片重均符合《中国药典》(2020年版)的相关规定。通过该研究发现利用大孔吸附树脂提取纯化的黄酮纯度较高,制成的片剂各项指标均符合相关规定,且工艺简单易行,可为黄酮片剂的推广和应用提供理论依据。 Using Actinidia arguta stems as the material,the crude extract of flavonoids was isolated and purified using AB-8macroporous adsorption resin.Different factors(sample concentration,sample flow rate,sample pH value,water washing volume,eluent concentration,eluent flow rate,eluent volume) were selected to investigate the effects of AB-8 macroporous adsorption resin on its adsorption and resolution ability.The orthogonal experimental method was used to investigate the effects of these factors on the adsorption and resolution ability of AB-8 macroporous adsorption resin,Explore the effects of flavonoid addition and the selection of different excipients on the preparation process of tablets,and conduct quality testing of the tablets.The best purification conditions obtained through the test are as follows:the concentration of the sample solution is 0.63 mg·mL^(-1),the flow rate of the sample solution is 3 mL·min^(-1),the pH value of the sample solution is 5.0,the water washing volume is 40 mL,the concentration of the eluent(ethanol) is 80%,the flow rate of the eluent is 3 mL·min^(-1),and the volume of the eluent is 30 mL.The purity of Flavonoid obtained through this scheme can reach 71.25%.The best formula of the tablet was optimized by single factor and orthogonal test.The results showed that the factors affecting the tablet were CBA(that is,the addition of carboxymethyl cellulose sodiumthe addition of starchthe flavonoid of Actinidia arguta stem),and the best formula was A2B1C1.Therefore,in the best formula of the tablet,the flavone content of Actinidia arguta stem is 38%,the starch content is 38%,the sodium carboxymethyl cellulose content is 8%,and the Mannitol content is 16%.The prepared tablet has a complete and smooth appearance,uniform color and no impurities,moderate hardness,and meets the relevant regulations of the Chinese Pharmacopoeia(2020 edition) in terms of disintegration time and tablet weight.Through this study,it was found that the purity of flavonoids extracted and purified using macroporous adsorption resin is high,and the various indicators of the prepared tablets comply with relevant regulations.The process is simple and feasible,which can provide a theoretical basis for the promotion and application of flavonoids tablets.
