摘要
目的探讨肌动蛋白结合蛋白2(TAGLN2)在人表皮生长因子受体2(HER2)阳性型和三阴性乳腺癌患者中的表达及临床意义。方法通过cBioPortal网站选取国际乳腺癌协会的分子分类数据库(METABRIC)中1904例浸润性乳腺癌患者的TAGLN2 mRNA表达数据和临床资料,以TAGLN2 mRNA表达值为标记,采用Kaplan-Meier法分析并计算TAGLN2 mRNA的cut-off值,根据该值将患者分为TAGLN2 mRNA高表达和TAGLN2mRNA低表达。比较不同受体表达状态乳腺癌患者TAGLN2 mRNA表达情况,比较HER2阳性型和三阴性乳腺癌患者的临床特征,比较不同TAGLN2 mRNA表达情况乳腺癌患者的生存情况,采用单因素和多因素Cox回归分析HER2阳性型和三阴性乳腺癌患者预后的影响因素。结果乳腺癌患者中TAGLN2 mRNA表达情况可能与雌激素受体(ER)表达、孕激素受体(PR)表达、HER2表达、HER2阳性(ER和PR阴性)、三阴性有关(P﹤0.01)。HER2阳性型和三阴性乳腺癌患者确诊年龄、肿瘤直径、肿瘤分期比较,差异均有统计学意义(P﹤0.05)。不同组织学分级三阴性乳腺癌患者TAGLN2 mRNA表达情况比较,差异有统计学意义(P﹤0.01)。HER2阳性型和三阴性乳腺癌患者的生存情况比较,差异无统计学意义(P﹥0.05);TAGLN2 mRNA低表达和TAGLN2 mRNA高表达HER2阳性型乳腺癌患者的生存情况比较,差异无统计学意义(P﹥0.05);TAGLN2 mRNA低表达和TAGLN2mRNA高表达三阴性乳腺癌患者的生存情况比较,差异无统计学意义(P﹥0.05)。多因素Cox回归分析显示,淋巴结转移情况是HER2阳性型乳腺癌患者预后的独立影响因素(P﹤0.01),确诊年龄和淋巴结转移情况均是三阴性乳腺癌患者预后的独立影响因素(P﹤0.01)。单因素分析显示,TAGLN2 mRNA表达情况不是HER2阳性型和三阴性乳腺癌患者预后的影响因素(P﹥0.05)。结论TAGLN2 mRNA表达情况不影响HER2阳性型和三阴性乳腺癌患者预后,可能不是这两种类型乳腺癌患者潜在的预后标志物。
Objective To investigate the expression and clinical significance of transgelin 2(TAGLN2)in human epidermal growth factor receptor 2(HER2)positive and triple negative breast cancer patients.Method The TAGLN2 mRNA expression data and clinical data of 1904 invasive breast cancer patients were obtained from Molecular Taxonomy of Breast Cancer International Consortium(METABRIC)database via the cBioPortal website.TAGLN2 mRNA expression level was set as the marker,and Kaplan-Meier method was used to analyze the cut-off value of TAGLN2 mRNA,and breast cancer patients were divided into TAGLN2 mRNA high expression and TAGLN2 mRNA low expression.The expression of TAGLN2 mRNA in breast cancer patients with different receptor expression states,the clinical characteristics of HER2 positive and triple negative breast cancer patients,and the survival of breast cancer patients with different TAGLN2 mRNA expression were compared.Univariate and multivariate Cox regression model were used to analyze the prognostic factors of HER2 positive and triple negative breast cancer patients.Result TAGLN2 mRNA expression in breast cancer patients was possibly related to the expression of estrogen receptor(ER),progesterone receptor(PR)and HER2,HER2 positive(ER and PR negative),and triple negative(P<0.01).There were significant differences in diagnosis age,tumor diameter and tumor stage between HER2 positive and triple negative breast cancer patients(P<0.05).There was significant difference for TAGLN2 mRNA expression in patients with triple negative breast cancer of different histological grades(P<0.01).There was no significant difference in survival between HER2 positive and triple negative breast cancer patients(P>0.05).There was no significant difference in survival of HER2 positive breast cancer patients with low expression of TAGLN2 mRNA and high expression of TAGLN2 mRNA(P>0.05).There was no significant difference in survival of triple negative breast cancer patients with low expression of TAGLN2 mRNA and high expression of TAGLN2 mRNA(P>0.05).Multivariate Cox regression analysis showed that lymph node metastasis was independent prognostic factor for HER2 positive breast cancer patients(P<0.01),and age at diagnosis and lymph node metastasis were independent prognostic factors for triple negative breast cancer patients(P<0.01).Univariate analysis showed that TAGLN2 mRNA expression was not prognostic factor for HER2 positive and triple negative breast cancer patients(P>0.05).Conclusion TAGLN2 mRNA did not affect the prognosis of HER2 positive and triple negative breast cancer patients,and may not be a potential prognostic marker for HER2 positive and triple negative breast cancer patients.
