摘要
由于人类活动干扰及自然环境变化,东北马鹿(Cervus elaphus xanthopygus)的生存面临诸多问题。为掌握东北马鹿遗传多样性现状,进而为马鹿种群的健康状况提供评估依据,利用线粒体Cyt b基因和微卫星2种分子标记对内蒙古大兴安岭南麓高格斯台罕乌拉国家级自然保护区内的东北马鹿进行种群遗传多样性评估。结果表明:246份新鲜马鹿粪便隶属于182只个体;微卫星标记表明平均期望杂合度为(0.729±0.016),平均观测杂合度为(0.768±0.029),线粒体标记表明单倍型多样性为(0.347±0.035),核苷酸多样性为(0.248±0.025)%,种群存在较高比例的稀有单倍型(33.33%)。该地区马鹿种群遗传多样性处于中等偏低水平,未来可能存在单倍型丢失的风险。因此,为避免出现遗传多样性下降的风险,建议该地区加强对马鹿种群的管理与保护,并促进与邻近区域马鹿个体的基因交流。
Due to the disturbance of human activities and changes in the natural environ⁃ment,the survival of red deer(Cervus elaphus xanthopygus)is facing many problems.To understand the current status of the genetic diversity of red deer and to provide a basis for assessing the health status,we evaluated the genetic diversity of red deer in the Gaogesitai-Hanwula National Nature Reserve in the southern of the Greater Khingan Mountains in Inner Mongolia using mitochondrial Cyt b and microsatellite markers.Results showed that 246 fresh feces belonged to 182 individuals.Microsatellite makers showed that the average expected heterozygosity(H_(e))was(0.729±0.016),and the average observed heterozygosity(H_(o))was(0.768±0.029).Mitochondrial marker showed that the haplotype diversity(H_(d))was(0.347±0.035),and the nucleotide diversity(Pi)was(0.248±0.025)%.There was a large proportion of rare haplotypes(33.33%)in this population.Overall,the population genetic diversity of red deer in this region was at a low to medium level.Therefore,to avoid the risk of genetic diversity decline in the future,it is recom⁃mended to strengthen the management and protection of red deer in this region,and there is a need to promote gene ex⁃change with red deer individuals in neighboring regions.
