电针肾俞穴对甲状腺素加负透镜诱导性近视豚鼠屈光参数和血清激素的影响

Effect of electroacupuncture on refractive parameters and serum hormones at Shenshu acupoint(BL 23)in thyroxine plus lens-induced myopia of guinea pigs

摘要 目的 观察电针肾俞穴对甲状腺素诱导性近视豚鼠的屈光参数和血清激素的影响。方法 选取2周龄三色健康短毛豚鼠50只,随机分为正常组、近视组、模型组、假穴组和电针组5组,每组10只豚鼠。近视组豚鼠右眼配戴-6.00 D透镜诱导近视,模型组、假穴组和电针组动物右眼配戴-6.00 D透镜诱导近视的同时,每日腹腔注射左旋甲状腺素钠0.05 mg·kg^(-1)·d^(-1)构建甲状腺功能亢进(简称甲亢)性近视模型;电针组于造模成功后(造模后第3周)开始电针肾俞穴治疗,持续4周;假穴组电针臀部假穴。正常组不做任何处理。观察并记录各组豚鼠造模后不同时间的一般情况、体重和肛温变化;同时利用检影镜和A超检测造模后0周、2周和6周时动物屈光度和眼轴长度。利用ELISA法检测造模后2周和6周各组豚鼠血清游离三碘甲状腺原氨酸(FT3)、游离甲状腺素(FT4)、促甲状腺素(TSH)和雌二醇(E2)水平变化。结果 造模后2周,与正常组比较,近视组、模型组、假穴组和电针组豚鼠的屈光度均显著下降(均为P<0.05);与近视组和正常组比较,模型组、假穴组和电针组豚鼠呈身体消瘦、烦躁不安、毛发枯槁、少尿和大便干燥等甲亢特征,且体重显著下降、肛温显著升高,血清激素FT3、FT4和E2水平均显著升高,TSH水平显著下降,差异均有统计学意义(均为P<0.05)。造模后6周(电针4周),与模型组和假穴组和比较,电针组豚鼠屈光度显著上升(P<0.05);身体健壮、性情温和、毛发顺滑有光泽、大小便正常;体重显著升高、肛温显著下降,血清激素FT3、FT4和E2水平均显著下降,TSH水平显著升高(均为P<0.05)。结论 电针肾俞穴可以显著抑制甲亢性近视豚鼠近视的进展,同时改善豚鼠体重、体温等甲亢症状,并使血清中FT3、FT4、E2和TSH等激素水平恢复平衡。 Objective To investigate the effects of electroacupuncture(EA)at shenshu acupoint(BL 23)on refractive parameters and serum hormones in guinea pigs with thyroxine-induced myopia.Methods Fifty 2-week-old healthy tricolor short-haired guinea pigs were selected and divided into 5 groups randomly:normal group,myopic group,model group,sham acupuncture(SA)group,and EA group,with 10 guinea pigs in each group.All guinea pigs in the myopic,model,SA,and EA groups wore a-6.00 D lens in the right eye to induce myopia.In addition,guinea pigs in the model,SA,and EA groups were intraperitoneally injected with 0.05 mg·kg^(-1)·d^(-1) L-thyroxine sodium daily to construct hyperthyroidism-associated myopia models.After 2 weeks,guinea pigs in the EA and SA groups were given EA at BL 23 and pseudo-acupuncture points respectively for 4 weeks.Guinea pigs in the normal group received no treatment.The general condition,body weight,and anal temperature of guinea pigs at different time points after modeling were monitored and recorded.The diopter and axial length were measured via retinoscopy and A-ultrasound imaging at 0,2,and 6 weeks after modeling.Serum-free triiodothyronine(FT3),free thyroxine(FT4),thyrotropin(TSH),and estradiol(E2)levels were measured via enzyme-linked immunosorbent assay at 2 and 6 weeks after modeling.Results Two weeks after modeling,the diopter of guinea pigs in the myopic,model,SA,and EA groups decreased significantly compared with the normal group(all P<0.05).Compared with the myopic and normal groups,the guinea pigs in the model,SA,and EA groups showed hyperthyroidism features such as body wasting,restlessness,withered hair,oliguria,and dry stools.Additionally,their body weight and TSH levels decreased significantly,while their anal temperature,FT3,FT4,and E2 levels increased significantly(all P<0.05).Compared with the model and SA groups,the diopter of guinea pigs in the EA group increased significantly six weeks after modeling(4 weeks after EA)(P<0.05);in addition,they became robust and mild-mannered,with a smooth and shiny coat and normal bowel movements;their body weight and TSH levels increased significantly,and their anal temperature,FT3,FT4,and E2 levels decreased significantly(all P<0.05).Conclusion EA at BL 23 can significantly inhibit the progression of hyperthyroidism\|associated myopia in guinea pigs,improve their body weight and temperature,and restore the balance of FT3,FT4,E2,and TSH levels.
