摘要
目的揭示齐墩果酸(oleanolic acid,OA)对免疫缺陷小鼠肠道免疫功能的影响。方法选取32只8周龄雌性昆明小鼠随机分为正常组、模型组、OA低剂量组与OA高剂量组。模型组与OA各剂量组连续5 d、1次/d腹腔注射环磷酰胺(80 mg/kg)制备免疫缺陷动物模型,正常组腹腔注射相应体积生理盐水。OA低剂量组(5 mg/kg)、OA高剂量组(25 mg/kg)第6天起1次/d灌胃给予OA(用2%Tween 80制成混悬液),正常组与模型组灌胃给予相应体积2%Tween 80,连续给药28 d。将正常组、模型组、OA低剂量组与OA高剂量组动物实施安乐死后摘除胸腺和脾脏,分析各组动物胸腺指数与脾脏指数,通过流式细胞术分析与比较各组动物肠系膜淋巴结T淋巴细胞亚群,通过酶联免疫吸附试验(ELISA)分析与比较各组动物肠黏膜分泌型免疫球蛋白A(secretary immunoglobulin A,sIgA)的水平。结果模型组动物胸腺指数(0.513±0.07)与脾脏指数(2.059±0.265)均显著低于正常组动物(P<0.05);OA高剂量组动物胸腺指数为(0.690±0.027),脾脏指数为(2.441±0.257),均显著高于模型组动物(P<0.05);OA高剂量组动物肠系膜淋巴结的CD3^(+)CD4^(+)T淋巴细胞占全部淋巴细胞的百分比为(42.70±5.31)%,CD3^(+)CD8^(+)T淋巴细胞占全部淋巴细胞的百分比为(18.33±1.90)%,均显著高于模型组动物(P<0.05);OA低剂量组动物肠系膜淋巴结的CD3^(+)CD8^(+)T淋巴细胞占全部淋巴细胞的百分比(16.48±3.80)%,显著高于模型组动物(P<0.05);OA高剂量组动物的肠黏膜sIgA含量为(1.38±0.38)ng/mL,显著高于模型组动物(P<0.05)。结论OA能够增加免疫缺陷小鼠胸腺指数与脾脏指数,增加肠系膜淋巴结T淋巴细胞的数量,促进肠黏膜sIgA的产生。OA对小鼠肠道免疫功能损伤具有修复作用。
Objective To investigate the effect of oleanolic acid(OA)on intestinal immune function of immunodeficient mice.Methods Thirty-two 8-week-old female Kunming mice were randomly divided into normal group,model group,OA low-dose group and OA high-dose group.The model group and OA groups were intraperitoneally injected with cyclophosphamide(80 mg/kg)once a day for 5 consecutive days,to prepare immunodeficiency animal models.The normal group was intraperitoneally injected with the corresponding volume of normal saline.OA low dose group(5 mg/kg)and OA high dose group(25 mg/kg)were intragastrically administered with OA(2%Tween 80 suspension)once a day from the 6th day,and the normal group and the model group were intragastrically administered with the corresponding volume of 2%Tween 80 for 28 days.After euthanasia,the immune organ indexes of animals in each group were analyzed,and the T lymphocyte subsets of mesenteric lymph nodes of animals in each group were analyzed and compared by flow cytometry.The levels of secretory immunoglobulin A(sIgA)in intestinal mucosa were analyzed and compared by enzyme-linked immunosorbent assay(ELISA).Results The thymus index(0.513±0.07)and spleen index(2.059±0.265)of mice in the model group were significantly lower than those in the normal group(P<0.05).The thymus index of mice in the high-dose OA group was(0.690±0.027),and the spleen index was(2.441±0.257),which were significantly higher than those of the model group(P<0.05).In the OA high-dose group,the percentage of CD3^(+)CD4^(+)T lymphocytes in the mesenteric lymph nodes was(42.70±5.31)%and the percentage of CD3^(+)CD8^(+)T lymphocytes was(18.33±1.90)%,which was significantly higher than that of the model group(P<0.05).The percentage of CD3^(+)CD8^(+)T lymphocytes in the mesenteric lymph nodes of the low-dose OA group was(16.48±3.80)%,which was significantly higher than that of the model group(P<0.05).The content of sIgA in intestinal mucosa of OA high dose group was 1.38±0.38(ng/mL),which was significantly higher than that of model group(P<0.05).Conclusion OA can increase the thymus index and spleen index of immunodeficiency mice,increase the number of T lymphocytes in mesenteric lymph nodes,and promote the production of sIgA in intestinal mucosa.OA has a repairing effect on intestinal immune function injury in mice.
