白藜芦醇对缺氧复氧致肾小管上皮细胞内质网应激和炎性损伤的影响 被引量:3

Effects of Resveratrol on Endoplasmic Reticulum Stress and Inflammatory Injury of Renal Tubular Epithelial Cells Induced by Hypoxia Reoxygenation

摘要 目的 探讨白藜芦醇对缺氧复氧致肾小管上皮细胞内质网应激和炎性损伤的影响。方法 将HK-2细胞分为对照组、模型组、白藜芦醇组,其中对照组放于37℃、95%空气及5%CO_(2)的培养箱中培养;模型组按照缺氧复氧处理方法培养;白藜芦醇组在缺氧复氧处理时换成含50 mg/L白藜芦醇的培养液,按照缺氧复氧处理方法培养。采用DCFH-DA检测细胞活性氧水平;采用流式细胞仪测定细胞凋亡率;采用Western blot法测定内质网应激蛋白糖调节蛋白78(GRP78)和C/EBP同源蛋白(CHOP)表达;采用ELISA法测定肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)水平。结果 模型组细胞活性氧表达明显增强,而白藜芦醇组细胞活性氧表达降低。模型组和白藜芦醇组细胞凋亡率高于对照组,而白藜芦醇组细胞凋亡率低于模型组(P<0.05)。模型组和白藜芦醇组GRP78和CHOP表达高于对照组,而白藜芦醇组GRP78和CHOP表达低于模型组(P<0.05)。模型组和白藜芦醇组TNF-α和IL-1β水平高于对照组,而白藜芦醇组TNF-α和IL-1β水平低于模型组(P<0.05)。结论 白藜芦醇可减轻缺氧复氧致肾小管上皮细胞凋亡,降低缺氧复氧致肾小管上皮细胞内质网应激和炎症因子水平,减轻炎性损伤。 Objective To investigate the effects of resveratrol on endoplasmic reticulum stress and inflammatory injury of renal tubular epithelial cells induced by hypoxia reoxygenation.Methods HK-2 cells were divided into control group,model group and resveratrol group.The control group was cultured in the incubator of 37℃,95%air and 5%CO2.The model group was cultured according to the method of hypoxia reoxygenation.The resveratrol group was cultured in the medium containing 50 mg/L resveratrol during hypoxia reoxygenation.DCFH-DA was used to detect the levels of reactive oxygen species.Flow cytometry was used to detect the apoptosis rate.Western blot was used to detect the expressions of endoplasmic reticulum stress protein GRP78 and CHOP.ELISA was used to detect the levels of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β).Results The expression of reactive oxygen species(ROS)in the model group was significantly increased,while that in the resveratrol group was decreased.The apoptosis rate of the model group and the tresveratrol group was higher than that of the control group,while the apoptosis rate of the resveratrol group was lower than that of the control group(P<0.05).The expressions of GRP78 and CHOP in the model group and the resveratrol group were higher than those in the control group,while the expressions of GRP78 and CHOP in the resveratrol group were lower than those in the model group(P<0.05).The levels of TNF-αand IL-1βin the model group and the resveratrol group were higher than those in the control group,while the levels of TNF-αand IL-1βin the resveratrol group were lower than those in the model group(P<0.05).Conclusion Resveratrol can reduce the apoptosis of renal tubular epithelial cells induced by hypoxia and reoxygenation,the endoplasmic reticulum stress and inflammatory factor levels of renal tubular epithelial cells induced by hypoxia and reoxygenation,and alleviate the inflammatory injury.
