摘要
研究适用于烟草及烟草制品总氮测定的消化工艺参数,寻找最佳催化剂配方和最佳消化温度、时间。采用正交实验进行了催化剂配方优选,文献法进行了消化温度、时间的初筛,分析总结进行了复筛,研究了总氮消化工艺参数,并选取典型烟草及烟草制品进行验证。结果表明:催化剂对消化结果的影响为:硫酸钾>硫酸>硫酸铜。其中,硫酸钾与硫酸和硫酸铜的比例用量上应达到平衡,硫酸钾用量不宜超过3 g。以样品量0.1 g、硫酸钾2 g、硫酸铜0.2 g、硫酸5 mL的催化剂配方在200℃(30 min)+420℃(130 min)的消化温度、时间条件下样品总氮含量达到真实值,较常规消化工艺参数时间缩短约2.33 h,检测效率提升87.5%。研究得到的消化工艺参数,操作安全,绿色高效,适用于各种来源及生产工艺的烟草及烟草制品。
To study the digestion parameters for the determination of total nitrogen(TN)in tobacco and tobacco products,and to search for the optimal catalyst formulation,dosage,digestion temperature and time.The orthogonal experiment was used to optimize catalyst formula,literature method for preliminary screening for digestion temperature and time,compound screening digestion of temperature and time were analyzed and summarized,Furthermore the typical tobacco and tobacco products were selected for verification.①the effect of catalyst on digestion was potassium sulfate>sulfuric acid>copper sulfate,and there was obvious interaction between the levels of each factor;②the content of total nitrogen reached the true value under the catalyst formulation of 200℃(30 min)+420℃(130 min)with the 0.1 g sample,2 g potassium sulfate,0.2 g copper sulfate and 5 mL sulfuric acid,The time was shortened by 2.33 h compared with the conventional methods,and the detection efficiency was improved by 87.5%.The digestive parameters obtained in this study are safe,green and efficient,and suitable for tobacco and tobacco products of various sources and production processes.
