摘要
目的:探讨染色体9p21单核苷酸多态性与新疆地区不同民族冠心病及西北燥证的相关性。方法:根据是否罹患西北燥证,分别收集维吾尔族燥证、非燥证冠心病患者各35例,汉族燥证、非燥证冠心病患者各35例,填写调查量表,比较冠心病燥证与非燥证患者病情积分、症状积分;采用基因测序法检测多态性,对基因突变位点进行精确的定位。结果:冠心病燥证组病情积分、症状积分均高于非燥证组(P<0.01);rs1333049、rs1333042、rs1333048、rs3217992、rs4977574、rs10757278位点基因型及等位基因在冠心病燥证和非燥证患者中的分布差异无统计学意义;在燥证组中,rs1333040的不同基因型,rs1333040、rs2383207位点的不同等位基因在维吾尔族组和汉族组中分布有显著性差异(P<0.05),与汉族燥证组比较,rs1333040位点CC基因型,rs1333040位点C等位基因及rs2383207位点A等位基因更易出现在维吾尔族燥证组中(P<0.05)。结论:西北燥证可能是新疆地区罹患冠心病患者人群的重要临床伴随证候,燥邪可能是影响新疆地区冠心病的发病及病情加重的重要因素之一;位点rs1333040、 rs2383207的多态性与新疆地区维吾尔族人群罹患冠心病伴西北燥证患者的易感性相关。
Objective:To investigate the association between chromosome 9p21 single nucleotide polymorphism and coronary heart disease(CHD)and northwest dryness syndrome in Uygur and Han populations in Xinjiang.Methods:According to whether they suffer from northwest dryness syndrome,35 cases of Uygur dryness syndrome and 35 cases of Han dryness syndrome and 35 cases of non-dryness syndrome were collected.Questionnaire was filled out to compare the disease scores and symptom scores of patients with dryness syndrome and non-dryness syndrome of CHD.Gene sequencing was used to detect polymorphisms and locate gene mutation sites accurately.Results:The disease score and symptom score of dryness syndrome group were higher than those of non-dryness syndrome group(P<0.01).The genotypes and alleles of rs1333049,rs1333042,rs1333048,rs3217992,rs4977574,rs10757278 were not significantly different in patients with dryland syndrome and non-dryland syndrome of CHD.In dryness syndrome group,the distribution of different genotypes of rs1333040,different alleles of rs1333040 and rs2383207 were statistically different between the Uygur and Han groups(P<0.05).Compared with the Han dryness syndrome group,the CC genotype of rs1333040 locus,C allele of rs1333040 locus and A allele of rs2383207 locus were more likely to appear in the dryness syndrome group(P<0.05).Conclusion:Northwest dryness syndrome may be an important clinical concomitant syndrome in patients with CHD in Xinjiang,and dryness may be one of the important factors affecting the incidence and aggravation of CHD in Xinjiang.The polymorphisms of rs1333040 and rs2383207 are associated with the susceptibility of patients with CHD and northwest dryness syndrome in Uygur population in Xinjiang.
