摘要
目的探讨淫羊藿苷对心力衰竭血清处理Dahl盐敏感大鼠骨髓源性树突状细胞(dendritic cells,DCs)的影响,为淫羊藿苷治疗心力衰竭提供实验依据。方法将50只SPF级5周龄雄性Dahl盐敏感大鼠及10只同龄雄性Dahl盐抵抗大鼠喂养至18周龄后麻醉制备心力衰竭血清与正常血清;分离Dahl盐敏感大鼠DCs,将DCs分为5组,模型组(心力衰竭血清),模型组+维生素D组(1×10^(-8)mol),模型组+淫羊藿苷高、中、低剂量组(40、20、10μmol/L),取Dahl盐抵抗大鼠DCs作为对照组(正常血清)。流式细胞仪检测6组DCs表面共刺激分子CD40、CD80、CD86、CD11b、MHC-II的表达情况;RT-PCR和Western-blot检测不同剂量淫羊藿苷对DCs维生素D受体(vitamin D receptor,VDR)、1α-羟化酶(CYP27B1)、24-羟化酶(CYP24A1)mRNA和蛋白的表达情况。结果淫羊藿苷与维生素D的治疗结果趋势一致,均可降低共刺激分子CD40、CD80、CD86及MHC-II的表达,升高CD11b的表达,显著上调VDR mRNA与CYP27B1 mRNA的表达和它们的蛋白相对表达量,下调CYP24A1 mRNA和蛋白相对表达量。结论淫羊藿苷与维生素D能够抑制DCs的分化与成熟,且淫羊藿苷与维生素D可以通过升高DCs VDR,CYP27B1的表达,降低CYP24A1的表达来有效治疗心力衰竭。
Objective To investigate the effect of icariin on the bone marrow derived dendritic cells(DCs)in Dahl salt-sensitive rats with heart failure serum,and provide experimental basis for icariin treating heart failure.Methods Fifty SPF-grade 5-week-old male Dahl salt-sensitive rats and 10 Dahl salt-resistant rats of the same age were fed to 18 weeks of age and anesthetized to prepare heart failure serum and normal serum.DCs of Dahl salt-sensitive rats were separated and divided into five groups:model group(heart failure serum),model group+vitamin D group(1×10^(-8)mol),model group+icariin high and low dose groups(40,20,10μmol/L),and the DCs of Dahl salt-resistant rats were taken out as the control group(normal serum).The expressions of co-stimulatory molecules CD40,CD80,CD86,CD11b and MHC-II on the surface of DCs in the six groups were detected by flow cytometry.RT-PCR and Western-blot were used to detect the effects of different doses of icariin on the expressions of vitamin D receptor(VDR),1α-hydroxylase(CYP27B1),24-hydroxylase(CYP24A1)mRNA and protein in DCs.Results The treatment results of icariin and vitamin D showed the same trend.Both of cariin and vitamin D reduced the expressions of co-stimulatory molecules CD40,CD80,CD86 and MHC-II and increased the expression of CD11b,significantly up-regulated the expressions of VDR mRNA and CYP27B1 mRNA and their relative protein expression,and down-regulated the relative expressions of CYP24A1 mRNA and protein.Conclusion Icariin and vitamin D can inhibit the differentiation and maturation of DCs,and icariin and vitamin D can effectively treat heart failure by increasing the expressions of VDR and CYP27B1 and decreasing the expression of CYP24A1 in DCs.
